A new class of drugs is now utilized for in vivo diagnoses and therapy of many widespread diseases. In these pharmacological and diagnostic preparations the active substance is conjugated with a vector which transports the drug to specific biological targets. Monoclonal antibodies are the most commonly used vectors: estimation of their permeability through multilayer and unilayer biomembranes is an important step in the analysis of efficiency of vector drugs.

An approach is proposed for measuring the binding constant (Kb) for monoclonal antibodies (MA) interacting with an immobilized antigen in indirect ELISA. This approach allows the measurement of optical density (A405) in the peroxidase reaction initiated by the conjugate at different concentrations (C0) of antibodies. Using the Scatchard plots, the dependence of A405/C0 = f (A405) for the whole range of MA concentrations was examined, and the tangential of the slope (tg alpha = Kb) of the linear portion of the antigen molecule was calculated.

Examination of children with chronic glomerulonephritis by means of using immunological indicators of the first level enabled a direct correlation (r = 60) to be established between the relative count of venous blood lymphocytes (the mean values of several studies) and the disease gravity. A regression equation is depicted, making it possible to identify a group at risk for a possible grave course of disease among patients with chronic glomerulonephritis. The relative count of T lymphocytes is the least dynamic and informative indicator for disease forecasting.

The enzymatic activity of blood leukocytes was studied in rats with Masugi nephritis by means of the quantitative cytochemical method. Nephrotoxin injection during first hours and days did not produce specific changes in the enzymatic activity. Maximum changes were observed on the 8-th day when the elevation of the activity of dehydrogenase lymphocytes and neutrophils acid phosphatase was observed at the same time with most pronounced histological changes in the kidneys, the decrease of the complement titer and the presence of nephrotoxin and autoglobulin in the renal glomeruli.
