Article Synopsis
  • The pachysandra alkaloids in Sarcococca ruscifolia have shown effectiveness against liver cancer but their structure and mechanisms are not fully understood.
  • Researchers synthesized twenty-three new derivatives of these alkaloids, resulting in compounds that were more potent and less toxic against HepG2 liver cancer cells than natural versions.
  • The best-performing compound, 6k, not only significantly inhibited cancer cell growth but also affected cancer cell migration and induced cell death by influencing the JAK2/STAT3 signaling pathway, indicating its potential as a treatment for hepatocellular carcinoma.

Objective: The research intended to probe the connection between the risk of stroke and serum vitamin D levels.

Methods: Three electronic databases (Cochrane Library, EMBASE, PubMed) were searched according to the subject terms from inception until July 29, 2022, and retrieved researches were screened on the basis of inclusion and exclusion criteria. Two investigators conducted the quality assessment and data extraction.

Seven undescribed abietane diterpenoids [abietamethinols A-G (1-7)] were isolated from the twigs and leaves of Isodon amethystoides. Their structures were elucidated on the basis of spectroscopic methods including 2D NMR, and they were further confirmed by X-ray crystallographic data. Lophanic acid was considered as the precursor of 1-7 in the biosynthesis pathway hypothesis.

Oridonin is one of the -kaurane diterpenes that have been studied extensively for various bioactivities. In an effort to expand natural scaffold-based library as anticancer agents, we have designed and synthesised a number of novel oridonin derivatives and evaluated their bioactivities on a panel of human cancer cell lines (HCT116, A375, MCF-7, HepG2, and A549). Compound bearing a 4-fluorophenyl moiety was found to be the most active compound with an IC value of 0.

Two undescribed -kaurene diterpenes, named guidongnins I () and J (), were isolated from the medicinal plant . Compound was determined to contain an unprecedented 23 carbons in the skeleton by bearing an extra isopropyl group at C-17 out of the diterpenoid parent structure, and compound was the first example of 6,7--7,20-olide--kaurenes with two fused-tetrahydrofuran rings formed between C-6 and C-19/C-20 through oxygen bridges. Their structures, including their absolute configurations, were determined using the analyses of the spectroscopic and X-ray diffraction data.

Isodon amethystoides (Lamiaceae) is a popular plant in folk medicine in the southern provinces of China. Our phytochemical investigation of the twigs and leaves of this plant led to the discovery of five new diterpenoids with isopimarane and 3,4-seco isopimarane scaffolds [isoamethinols A-E (1-5)], along with the known compound 3,4-seco isopimara-4(18),7,15-triene-3-oic acid methylester (6). The chemical structures of these compounds, including the absolute configurations of the new diterpenoids, were determined by comprehensive spectroscopic analyses and single crystal X-ray diffraction measurements.

Warburg effect of aerobic glycolysis in hepatic M1 macrophages is a major cause for metabolic dysfunction and inflammatory stress in non-alcoholic fatty liver disease (NAFLD). Plant-derived triterpene celastrol markedly inhibited macrophage M1 polarization and adipocyte hypertrophy in obesity. The present study was designed to identify the celastrol-bound proteins which reprogrammed metabolic and inflammatory pathways in M1 macrophages.

We previously found that sulfur fumigation, a commonly used controversial method for the post-harvest handling of ginger, induces the generation of a compound in ginger, which was speculated to be a sulfur-containing derivative of 6-shogaol based on its mass data. However, the chemical and biological properties of the compound remain unknown. As a follow-up study, here we report the chemical structure, systemic exposure, and anticancer activity of the compound.

Article Synopsis
  • Three new diterpenes (fladins B, C, and D) were extracted from Chinese medicinal plants, and their chemical structures were confirmed using advanced techniques like X-ray crystallography.
  • These compounds have a unique structure formed through a rearrangement process, making them the first known isopimarane diterpenes with a specific lactone feature.
  • While most of the isolated compounds showed no significant cytotoxic or antiviral activities, one compound demonstrated an 81% inhibitory effect against Ebola, indicating potential for further development as an antiviral agent.

Increasing evidence shows that autophagy impairment is involved in the pathogenesis and progression of neurodegenerative diseases including Parkinson's disease (PD). We previously identified a natural alkaloid named corynoxine B (Cory B) as a neuronal autophagy inducer. However, its brain permeability is relatively low, which hinders its potential use in treating PD.

A phytochemical investigation of the leaves of the medicinal plant led to the isolation of the two new degraded abietane lactone diterpenoids rubesanolides F () and G (). Their structures were elucidated based on the analyses of the HRESIMS and 1D/2D NMR spectral data, and their absolute configurations were determined by ECD spectrum calculations and X-ray single crystal diffraction methods. Compounds and , with a unique γ-lactone subgroup between C-8 and C-20, were found to form a carbonyl carbon at C-13 by removal of the isopropyl group in an abietane diterpene skeleton.

Arylnaphthalene lignans (ANLs) were known to have axial chirality due to the biphenyl skeleton with hindered rotation at the single bond. However, the stable ANL atropisomers have not been isolated from nature until the present study. Phytochemical separation of the methanol extract of the stems and barks of led to the isolation of 11 ANL glycosides including four pairs of new atropisomers with stable confirmations at room temperature.

Introduction: The effect of hydrogen sulfide (H2S) on global cerebral ischemia remains partially understood. This study aimed to investigate the neuroprotective effect of sodium hydrosulfide (NaHS, a donor of H2S) post-conditioning and its underlying mechanism in a transient global cerebral ischemia (tGCI) model.

Materials & Methods: The tGCI rat model was established by the four-vessel occlusion method.

Article Synopsis
  • * Post-treatment with specific doses of NaHS significantly increased the survival of hippocampal neurons and improved their overall health after ischemia.
  • * The neuroprotective effects of NaHS may be linked to its ability to regulate the expression of potassium channels (Kv4.2 and KChIP3) in the brain, suggesting a potential therapeutic role for NaHS in ischemic injuries.
