Total proctocolectomy with ileal pouch anal anastomosis is the standard of care for patients with severe ulcerative colitis. We generated a cell-type-resolved transcriptional and epigenetic atlas of ileal pouches using scRNA-seq and scATAC-seq data from paired biopsy samples of the ileal pouch and the ileal segment above the pouch (pre-pouch) from patients (male=4, female=2), and paired biopsies of the terminal ileum and ascending colon from healthy individuals (male=3, female=3) serving as reference. Our study finds an additional population of absorptive and secretory epithelial cells within the pouch but not the pre-pouch.

The fallopian tube undergoes extensive molecular changes during the menstrual cycle and menopause. We use single-cell RNA and ATAC sequencing to construct a comprehensive cell atlas of healthy human fallopian tubes during the menstrual cycle and menopause. Our scRNA-seq comparison of 85,107 pre- and 46,111 post-menopausal fallopian tube cells reveals substantial shifts in cell type frequencies, gene expression, transcription factor activity, and cell-to-cell communications during menopause and menstrual cycle.

Among the various aflatoxin B1 (AFB1) assays, performing accurate detection is difficult because false positives and false negatives are frequent due to limited sensitivity, expensive equipment, or inadequate pretreatment during operation. Here, an "off-on" switch-type electrochemiluminescence (ECL) aptasensor armed with cobalt-sulfur quantum dots was encapsulated in hollow cobalt-layered double hydroxide nanocages as an enhanced luminescent probe (Co-LDH@QDs), and a ferrocene-modified aptamer (Fc-APT) was used as a luminescent quencher. In general, when Fc-APT was hybridized with complementary DNA modified with a DNA nanotetrahedron, electron transfer between ferrocene and Co-LDH@QDs was facilitated, leading to efficient quenching of the ECL intensity into an "off" state in the absence of AFB1.

Background: To investigate the association between periodontitis and inflammatory biomarkers, including systemic immune-inflammatory index (SII), neutrophil-to-lymphocyte ratio, platelet-to-lymphocyte ratio, and lymphocyte-to-monocyte ratio.

Methods: Our study comprised a cross-sectional analysis (an indirect evidence group and a periodontal health control group from January to October 2023) and a retrospective study (a direct evidence group and a non-maintenance group from January 2014 to March 2022). We analyzed demographic data, imaging measurements, and peripheral blood counts.

Article Synopsis
  • The N6-methyladenine (m6A) modification is key in eukaryotic mRNAs, playing significant roles in cancers like bladder cancer (BLCA).
  • This study investigates how the protein ZC3H13 influences m6A modification in BLCA, finding that ZC3H13 is lower in cancer tissues and inhibits cancer cell growth.
  • ZC3H13 helps stabilize PJA2 mRNA, leading to degradation of KSR1 and promoting autophagy, which together contribute to reduced BLCA progression.

Transcriptome-wide association studies (TWAS) help identify disease causing genes, but often fail to pinpoint disease mechanisms at the cellular level because of the limited sample sizes and sparsity of cell-type-specific expression data. Here we propose scPrediXcan which integrates state-of-the-art deep learning approaches that predict epigenetic features from DNA sequences with the canonical TWAS framework. Our prediction approach, ctPred, predicts cell-type-specific expression with high accuracy and captures complex gene regulatory grammar that linear models overlook.

Monitoring biomarkers secreted by cardiomyocytes is critical to evaluate anticancer drug-induced myocardial injury (MI). Cardiac troponin I (cTnI) is considered the gold standard biomarker for MI. Herein, an electrochemical aptasensor is engineered for cTnI detection based on lanthanide europium metal-organic frameworks (Eu-MOFs) and a hybridization chain reaction-directed DNAzyme strategy.

  • Insulin resistance (IR) is linked to many metabolic diseases, and the gut microbiota plays a key role in regulating IR by maintaining gut health.
  • A specific microorganism, Akkermansia muciniphila, helps keep the intestinal barrier intact, reduces inflammation, and improves insulin sensitivity, making its abundance important for preventing IR.
  • Research into A. muciniphila's effects on IR could lead to personalized medicine approaches that optimize gut health to combat metabolic disorders.

To analyze the efficacy of endoscopic CO₂ laser arytenoidectomy in treating bilateral vocal cord paralysis. Ninety-five patients who underwent endoscopic CO₂ laser arytenoidectomy for bilateral vocal cord paralysis at the First Affiliated Hospital of Naval Medical University(Changhai Hospital) of Shanghai from January 2009 to December 2022 were included in this study. Among them, 59 patients underwent endoscopic CO₂ laser arytenoidectomy as their first glottic enlargement surgery(Group A), while 36 patients, who had previously undergone two unsuccessful CO₂ laser-assisted posterior cordotomies, underwent endoscopic CO₂ laser arytenoidectomy as a subsequent surgery(Group B).

  • The study compares the effects of different surgical methods (staged resection, one-stage surgery with laryngeal stent, and simple CO₂ laser excision) on early glottic cancer involving the anterior commissure in 83 patients with T1-2N0M0 stage cancer.
  • Important outcomes assessed include primary tumor control, vocal function recovery measured by VHI-10 and MPT, and postoperative adhesion rates and degree of vocal cord adhesion.
  • Results showed that Group C (simple CO₂ laser excision) had a higher adhesion rate and poorer vocal function recovery compared to Groups A (staged resection) and B (one-stage with stent), indicating that methods involving more invasive procedures may lead

Objective: Fetal growth restriction (FGR) is a condition where fetuses fail to reach their genetic potential for growth, posing a significant health challenge for newborns. The aim of this research was to explore the efficacy of texture-based analysis of neurosonographic images in identifying FGR in fetuses, which may provide a promising tool for early assessment of FGR.

Methods: A retrospective analysis collected 100 intrauterine neurosonographic images from 50 FGR and 50 gestational age-appropriate fetuses.

Chelidonii herba is a traditional Chinese medicinal herb with effects including antispasmodic, analgesic, antitussive, and bronchodilator properties. Alkaloids are the main bioactive ingredients in Chelidonii herba. In this study, a two-dimensional nuclear magnetic resonance (Heteronuclear Singular Quantum Correlation, HSQC-2D-NMR) technique was employed to quantitatively analyze the total alkaloid content and three major active alkaloid monomers in Chelidonii herba from eleven different sources.

Epithelial cell adhesion molecule (EpCAM), which is overexpressed in breast cancer cells and participates in cell signaling, migration, proliferation, and differentiation, has been utilized as a biomarker for cancer diagnosis and therapeutic prognosis. Here, a dual-signal readout nonenzymatic aptasensor is fabricated for the evaluation of EpCAM at the level of three breast cancer cell lines. The central principle of this enzyme-free aptasensor is the use of double hook-type aptamers (SYL3C and SJ3C2)-functionalized magnetic iron oxide (FeO) as capture probes and quasi-CoFe prussian blue analogs (QCoFe PBAs) as nonenzymatic signal probes for colorimetric and electrochemical analysis.
