Objective: This study aimed to evaluate the effects of different cold acclimation strategies on exercise performance in male mice exposed to low-temperature environments.

Methods: Male mice were subjected to five distinct acclimation regimens over 8 weeks: immersion at 10 °C (10 °CI) or 20 °C (20 °CI), swimming at 10 °C (10 °CS), 20 °C (20 °CS), or 34 °C (34 °CS). During the first 2 weeks, the acclimation time progressively decreased from 30 min to 3 min per day, and the water temperatures were lowered from 34 °C to the target levels, followed by 6 weeks of consistent exposure.

Transient receptor potential melastatin 2 (TRPM2) is a non-selective cation channel that exhibits Ca permeability. The TRPM2 channel is expressed in various tissues and cells and can be activated by multiple factors, including endogenous ligands, Ca, reactive oxygen species (ROS) and temperature. Lactic acid (LA) is a powerful molecule as the metabolic driver in tumor microenvironments (TMEs). Inspired by its high intratumoral level (5-20 µmol g ), a novel treatment paradigm via the cascade release of H O and ·OH from the LA generated by tumor metabolism is developed for catalytic and pH-dependent selective tumor chemotherapy. By utilizing the acidity and overexpression of LA within the TME, the constructed lactate oxidase (LOD)-immobilized Ce-benzenetricarboxylic acid (Ce-BTC) metal organic framework enables the intratumoral generation of ·OH via a cascade reaction: 1) the in situ catalytic release of H O from LA by LOD, and 2) the catalytic production of ·OH from H O by Ce-BTC with peroxidase-like activity.
