The core component of the class III phosphatidylinositol 3-kinase complex, Beclin 1, takes part in different protein networks, thus switching its role from inducing autophagy to regulating autophagosomal maturation and endosomal trafficking. While assessed in neurons, astrocytes, and microglia, its role is far less investigated in myelinating glia, including Schwann cells (SCs), responsible for peripheral nerve myelination. Remarkably, the dysregulation in endosomal trafficking is emerging as a pathophysiological mechanism underlying peripheral neuropathies, such as demyelinating Charcot-Marie-Tooth (CMT) diseases.

Liver transplantation (LT) associates with weight gain and metabolic complications. However, risk of eating disorders post-transplantation and factors influencing their onset remain poorly understood. This study aimed to fill this knowledge gap by characterizing the risk of having eating disorders or Orthorexia Nervosa (ON) according to the EAT-26, BES and Bratman screening questionnaires in 104 liver transplant recipients (mean age 62.

Chemotherapy can cause vascular and metabolic liver injury in patients with liver metastases, but scarce data are available. We aimed to (i) describe the prevalence of porto-sinusoidal vascular disorder (PSVD) among patients undergoing resection for liver metastases; and (ii) assess whether liver (LSM) and spleen stiffness measurements could diagnose PSVD and predict postoperative complications. This is a prospective single-center study enrolling consecutive patients undergoing hepatic resection for metastases at a tertiary center.

Dual hypothermic oxygenated perfusion (DHOPE) is increasingly being used to extend liver preservation to improve transplant logistics. However, little is known about its benefits in high-risk liver grafts. This study aimed to investigate whether prolonged DHOPE provides benefits other than improved logistics in all liver types.

Article Synopsis
  • Recent international guidelines have defined metabolic dysfunction-associated steatotic liver disease (MASLD), leading to a rise in its prevalence as a reason for liver transplantation (LT) in Italy between 2012 and 2022.
  • * An analysis of 1,941 patients with MASLD and 11,201 without MASLD revealed that while the prevalence of MASLD patients waiting for LT increased significantly, it did not independently affect patient survival post-transplant.
  • * Patients with non-HCC MASLD faced higher mortality rates on the waiting list but projected better 5-year transplant survival benefits than those without MASLD or HCC.

Article Synopsis
  • Scientists are studying how to tell if a donated liver is healthy enough to use for transplant by looking at special markers during a process called HOPE.
  • They collected samples from livers in 10 different centers across 7 countries and found that the levels of a marker called FMN can help predict if the liver will work well after being transplanted.
  • The study showed that FMN is better at predicting liver problems compared to older methods, making it a promising tool for doctors to decide which livers are suitable for transplant.

Article Synopsis
  • Liver transplantation (LT) was once thought not to work for a type of cancer called perihilar cholangiocarcinoma (pCCA), but a new method from the Mayo Clinic showed it could help if patients followed strict rules and took certain treatments first.
  • In a study of 22 liver transplant centers in Italy, 53 patients with pCCA were looked at, with some getting special treatment before the transplant and others not.
  • The results showed that patients who had the pre-treatment did better after 1, 3, and 5 years compared to those who didn’t, leading to talks about making new rules for how to treat this cancer better in Italy.

Purpose: Single large hepatocellular carcinoma >5cm (SLHCC) traditionally requires a major liver resection. Minor resections are often performed with the goal to reduce morbidity and mortality. Aim of the study was to establish if a major resection should be considered the best treatment for SLHCC or a more limited resection should be preferred.

Collagen VI (Col-VI) is an extracellular matrix protein primarily known for its bridging role in connective tissues that has been suggested to play a neuroprotective role. In the present study we report increased mRNA and protein expression of Col-VI in the hippocampus and cortex at a late stage of epileptogenesis in a post-status epilepticus (SE) model of epilepsy and in brain tissue from patients with epilepsy. We further present a novel finding that exposure of mouse hippocampal slices to Col-VI augments paired-pulse facilitation in Schaffer collateral-CA1 excitatory synapses indicating decreased release probability of glutamate.

Article Synopsis
  • * This study examined how pericytes function in elderly individuals and those with muscular dystrophies linked to collagen VI mutations, highlighting the challenges of aging and muscle degeneration.
  • * The research demonstrated that aging affects pericytes negatively, impairing their ability to help form blood vessels, while young patients with collagen VI issues showed some similar traits but retained a stronger ability to cope with oxidative stress and support blood vessel formation.

Article Synopsis
  • A new study evaluated long-term outcomes of liver transplants from donors treated with hypothermic oxygenated machine perfusion (HOPE) across 22 European centers, involving 1,202 transplant cases from 2012 to 2021.
  • The study showed high 1-, 3-, and 5-year death-censored graft survival rates for both donation after brain death (DBD) and donation after circulatory death (DCD), with low rates of primary non-function (PNF) and ischaemic cholangiopathy (IC).
  • The findings indicate that HOPE treatment effectively improves liver transplant outcomes, regardless of the donor's risk profile, and supports its incorporation into routine clinical practice.

Gut microbiota is responsible for essential functions in human health. Several communication axes between gut microbiota and other organs via neural, endocrine, and immune pathways have been described, and perturbation of gut microbiota composition has been implicated in the onset and progression of an emerging number of diseases. Here, we analyzed peripheral nerves, dorsal root ganglia (DRG), and skeletal muscles of neonatal and young adult mice with the following gut microbiota status: a) germ-free (GF), b) gnotobiotic, selectively colonized with 12 specific gut bacterial strains (Oligo-Mouse-Microbiota, OMM12), or c) natural complex gut microbiota (CGM).

Background: For many tumors, radiomics provided a relevant prognostic contribution. This study tested whether the computed tomography (CT)-based textural features of intrahepatic cholangiocarcinoma (ICC) and peritumoral tissue improve the prediction of survival after resection compared with the standard clinical indices.

Methods: All consecutive patients affected by ICC who underwent hepatectomy at six high-volume centers (2009-2019) were considered for the study.

Contamination by pharmaceuticals adversely affects the quality of natural water, causing environmental and health concerns. In this study, target drugs (oxazepam, OZ, 17-α-ethinylestradiol, EE2, and drospirenone, DRO), which have been extensively detected in the effluents of WWTPs over the past decades, were selected. We report here a new photoactive system, operating under visible light, capable of degrading EE2, OZ and DRO in water.
