In this work we investigated how the neurosteroid allopregnanolone can modulate learning and memory processes. For this purpose, we used ovariectomized (OVX) rats subcutaneously injected with oestradiol benzoate (E) alone or E and progesterone (P). Then, rats were injected in dorsal hippocampus with allopregnanolone or vehicle.

Ghrelin (Ghr) is a peptide produced peripherally and centrally. It participates in the modulation of different biological processes. In our laboratory we have shown that (a) Ghr administration, either intracerebroventricular or directly into the hippocampus enhanced memory consolidation in a step down test in rats (b) the effect of Ghr upon memory decreases in animals pretreated with a serotonin (5-HT) reuptake inhibitor, Fluoxetine, suggesting that Ghr effects in the hippocampus could be related to the availability of 5-HT.

Hemiparkinsonism induced by 6-hydroxydopamine (6-OHDA) injected in left corpus striatum is a recognized model of motor deficits in rats. Some reports concerning motor deficits indicate a favorable response to steroid administration in hemiparkinsonian animals. However, there is no much information regarding progesterone administration in relation to cognitive and affective dysfunctions.

Both peripheral innervation and nitric oxide (NO) participate in ovarian steroidogenesis. The aims of the work were (1) to investigate whether ganglionic noradrenergic (NE) and cholinergic (Ach) stimulus modify the ovarian steroids and NO release and (2) to examine the effect of those stimuli on the mRNA expression of 3beta-HSD and P450 aromatase in the ovary. The experiments were carried out using the ex vivo coeliac ganglion-superior ovarian nerve-ovary (CG-SON-O) system of rats in the first oestral cycle.
