Heparan sulfate (HS) proteoglycans are important regulators of cellular responses to soluble mediators such as chemokines, cytokines and growth factors. We profiled changes in expression of genes encoding HS core proteins, biosynthesis enzymes and modifiers during macrophage polarisation, and found that the most highly regulated gene was Sulf2, an extracellular HS 6-O-sulfatase that was markedly downregulated in response to pro-inflammatory stimuli. We then generated Sulf2 bone marrow chimeric mice and examined inflammatory responses in antigen-induced arthritis, as a model of rheumatoid arthritis.

Background: Testicular germ cell tumours (TGCTs) represent a clinical challenge; they are most prevalent in young individuals and are triggered by molecular mechanisms that are not fully understood. The origin of TGCTs can be traced back to primordial germ cells that fail to mature during embryonic development. These cells express high levels of pluripotency factors, including the transcription factor NANOG which is highly expressed in TGCTs.

In critically ill newborns, exposure to hypercapnia (HC) is common and often accepted in neonatal intensive care units to prevent severe lung injury. However, as a "safe" range of arterial partial pressure of carbon dioxide levels in neonates has not been established, the potential impact of HC on the neurodevelopmental outcomes in these newborns remains a matter of concern. Here, in a newborn Yorkshire piglet model of either sex, we show that acute exposure to HC induced persistent cortical neuronal injury, associated cognitive and learning deficits, and long-term suppression of cortical electroencephalogram frequencies.

Biomass burning is common in much of the world, and in some areas, residential wood-burning has increased. However, air pollution resulting from biomass burning is an important public health problem. A sampling campaign was carried out between May 2017 and July 2018 in over 64 sites in four sessions, to develop a spatio-temporal land use regression (LUR) model for fine particulate matter (PM) and wood-burning tracers levoglucosan and soluble potassium (K) in a city heavily impacted by wood-burning.

Understanding how tropical cyclones affect phytoplankton communities is important for studies on ecological variability. Most studies assessing the post-storm phytoplankton response rely on satellite observations of chlorophyll a concentration, which inform on the ocean surface conditions and the whole phytoplankton community. In this work, we assess the potential of the Massachusetts Institute of Technology marine ecosystem model to account for the response of individual phytoplankton functional types (PFTs, coccolithophores, diatoms, diazotrophs, mixotrophic dinoflagellates, picoeukaryotes, Prochlorococcus and Synechococcus) in the euphotic zone to the passage of Hurricane Fabian (2003) across the tropical and subtropical Sargasso Sea.

Article Synopsis
  • Estimating health loss trends in Peru from 1990 to 2019 is crucial for improving healthcare resource allocation and system performance, using data from the Global Burden of Disease study.
  • The Peruvian population rose to 33.9 million by 2019, with life expectancy at birth increasing from 69.2 to 80.3 years, largely due to a decrease in under-5 mortality and infectious diseases, while disability-adjusted life-years (DALYs) showed a decline in overall health burden.
  • There was a notable shift towards non-communicable diseases as the leading cause of health loss, with major contributors to DALYs in 2019 including neonatal disorders, lower respiratory infections, and lifestyle

Background: Arbovirus diseases such as dengue, Zika, and chikungunya are a public health threat in tropical and subtropical areas. In the absence of a vaccine or specific treatment, vector management (in this case the control of the primary vector Aedes aegypti) is the best practice to prevent the three diseases. A good understanding of vector behaviour, ecology, human mobility and water use can help design effective vector control programmes.

Clinical management of prostate cancer is challenging because of its highly variable natural history and so there is a need for improved predictors of outcome in non-metastatic men at the time of diagnosis. In this study we calculated the model score from the leading clinical multivariable model, PREDICT prostate, and the poor prognosis DESNT molecular subtype, in a combined expression and clinical dataset that were taken from malignant tissue at prostatectomy (n = 359). Both PREDICT score (p < 0.

Rheumatoid arthritis (RA)-associated interstitial lung disease (RA-ILD) is the most common pulmonary complication of RA, increasing morbidity and mortality. Anti-citrullinated protein antibodies have been associated with the development and progression of both RA and fibrotic lung disease; however, the role of protein citrullination in RA-ILD remains unclear. Here, we demonstrate that the expression of peptidylarginine deiminase 2 (PAD2), an enzyme that catalyzes protein citrullination, is increased in lung homogenates from subjects with RA-ILD and their lung fibroblasts.
