Article Synopsis
  • Polygenic risk scores (PRS) can help target colorectal cancer (CRC) screening for those at higher risk, but current versions are less effective for non-European populations.
  • Researchers combined data from Asian ancestry with European ancestry datasets to improve PRS accuracy, achieving better performance across different racial/ethnic groups.
  • The study suggests that adding more non-European data, particularly from Black/African American and Latinx/Hispanic populations, is essential for enhancing risk prediction and promoting equitable clinical practices.

Monocyte chemoattractant protein-1 (MCP-1/CCL2) is a potent chemotactic agent for monocytes, primarily produced by macrophages and endothelial cells. Significantly elevated levels of MCP-1/CCL2 were found in synovial fluids of patients with rheumatoid arthritis (RA), compared to osteoarthritis or other arthritis patients. Several studies suggested an important role for MCP-1 in the massive inflammation at the damaged joint, in part due to its chemotactic and angiogenic effects.

Self-reported type 2 diabetes mellitus (T2DM) is a risk factor for many cancers, suggesting its pathology relates to carcinogenesis. We conducted a case-cohort study to examine associations of hemoglobin A (HbA) and c-peptide with cancers associated with self-reported T2DM. This study was drawn from a prospective cohort of 32,383 women and men who provided blood specimens at baseline: c-peptide and HbA were assessed in 3,000 randomly selected participants who were cancer-free-at-baseline and an additional 2,281 participants who were cancer-free-at-baseline and subsequently diagnosed with incident colorectal, liver, pancreatic, female breast, endometrial, ovarian, bladder, or kidney cancers.

APOBEC mutational signatures SBS2 and SBS13 are common in many human cancer types. However, there is an incomplete understanding of its stimulus, when it occurs in the progression from normal to cancer cell and the APOBEC enzymes responsible. Here we whole-genome sequenced 342 microdissected normal epithelial crypts from the small intestines of 39 individuals and found that SBS2/SBS13 mutations were present in 17% of crypts, more frequent than most other normal tissues.

Article Synopsis
  • The study investigates how the location of colorectal tumors affects survival rates, particularly in relation to their molecular characteristics like microsatellite instability (MSI) and methylation patterns (CIMP).
  • A large dataset of over 13,000 colorectal cancer cases revealed significant survival trends based on tumor location, with non-MSI-high tumors showing better outcomes as they moved from the cecum to the sigmoid colon, while MSI-high tumors had worse outcomes in the same locations.
  • The findings emphasize the need for considering both tumor location and molecular features for more accurate prognostication in colon cancer, highlighting the potential of precision medicine in improving patient outcomes.

Article Synopsis
  • Giant tortoises were once widespread on various islands in the western Indian Ocean before humans arrived; this study investigates their diversity and biogeography using ancient DNA and other methods.
  • The research proposes that the ancestor of the extinct Mascarene tortoises migrated from Africa to now-submerged islands northeast of Madagascar during the Eocene, leading to multiple colonization events.
  • The study reveals that several giant and large tortoise species from Madagascar disappeared between 1000 and 600 years ago, highlighting how integrating ancient DNA with other data sources helps illuminate the history of island species.

Aims: Long-term retention of impacted third molars (wisdom teeth) is associated with plaque stagnation and the development of caries on the adjacent surface of the neighboring second molar. While caries and tooth loss are common outcomes of impaction, there is currently insufficient evidence to support the pre-emptive removal of asymptomatic wisdom teeth. Emerging evidence suggests that convergently growing impactions are associated with caries.

Article Synopsis
  • Tobacco smoking increases the risk of colorectal cancer, and certain genetic profiles may heighten this risk further.
  • A study involving over 33,000 colorectal cancer cases and nearly 44,000 controls identified specific genetic loci (on chromosomes 3p12.1, 6p21.33, and 8q24.23) that interact with smoking behaviors, potentially leading to a greater risk of developing the disease.
  • The research suggests that higher expression of specific genes is associated with a lower risk of colorectal cancer, emphasizing the role of these genetic factors in smoking-related cancer susceptibility and potential avenues for prevention.

A consequence of over 400 years of human exploitation of Galápagos tortoises (Chelonoidis niger ssp.) is the extinction of several subspecies and the decimation of others. As humans captured, killed, and/or removed tortoises for food, oil, museums, and zoos, they also colonized the archipelago resulting in the introduction of invasive plants, animals, and manipulated landscapes for farming, ranching, and infrastructure.
