Pannexin 1 (PANX1) forms cell-surface channels capable of releasing signaling metabolites for diverse patho-physiological processes. While inhibiting dysregulated PANX1 has been proposed as a therapeutic strategy for many pathological conditions, including inflammatory bowel disease (IBD), low efficacy, or poor specificity of classical PANX1 inhibitors introduces uncertainty for their applications in basic and translational research. Here, hit-to-lead optimization is performed and a naphthyridone, compound 12, is identified as a new PANX1 inhibitor with an IC of 0.

Statistical combinations of searches for charginos and neutralinos using various decay channels are performed using 139  fb^{-1} of pp collision data at sqrt[s]=13  TeV with the ATLAS detector at the Large Hadron Collider. Searches targeting pure-wino chargino pair production, pure-wino chargino-neutralino production, or Higgsino production decaying via standard model W, Z, or h bosons are combined to extend the mass reach to the produced supersymmetric particles by 30-100 GeV. The depth of the sensitivity of the original searches is also improved by the combinations, lowering the 95% C.

Article Synopsis
  • The immune synapse is a crucial structure for T cell activation, connecting T lymphocytes with antigen-presenting cells (APCs).
  • Research reveals that INPP5E, a protein associated with the sensory organelle called cilia, is concentrated at the immune synapse in Jurkat T-cells during specific types of T cell activation.
  • Silencing INPP5E hampers key signaling processes, affecting the proper function of TCR signaling and reducing the secretion of IL-2, an important immune response factor.

A test of CP invariance in Higgs boson production via vector-boson fusion has been performed in the H→γγ channel using 139  fb^{-1} of proton-proton collision data at sqrt[s]=13  TeV collected by the ATLAS detector at the LHC. The optimal observable method is used to probe the CP structure of interactions between the Higgs boson and electroweak gauge bosons, as described by an effective field theory. No sign of CP violation is observed in the data.

Formyl peptide receptor-like 1 inhibitor protein (FLIPr) is an immune evasion protein produced by Staphylococcus aureus, and FLIPr is a potential vaccine candidate for reducing Staphylococcus aureus virulence and biofilm formation. We produced recombinant lipidated FLIPr (rLF) to increase the immunogenicity of FLIPr and showed that rLF alone elicited potent anti-FLIPr antibody responses to overcome the FLIPr-mediated inhibition of phagocytosis. In addition, rLF has potent immunostimulatory properties.

Article Synopsis
  • Ulcerative colitis (UC) and Crohn's disease are forms of inflammatory bowel disease (IBD) caused by abnormal immune responses, and the study focuses on the less understood interactions among immune cells and the mechanisms behind the drug vedolizumab.
  • Researchers used a method called CITE-seq on immune cells from UC patients treated with vedolizumab and applied a computational tool (NicheNet) to study immune cell interactions and transcriptional changes.
  • Findings showed that UC patients who responded to vedolizumab had fewer T helper 17 (TH17) cells and different interaction patterns with other immune cells compared to nonresponders, suggesting that understanding cell communications could enhance treatments for IBD.

The microorganisms inhabiting our gastrointestinal tract are critical for human health. Chronic heavy alcohol use can modulate the composition and function of the gut microbiota, thereby exacerbating end-organ damage via the gut-brain axis and the gut-liver axis. In this review, we summarize the bacterial, fungal, and viral gut microbial compositional changes associated with alcohol use and alcohol-associated liver disease and discuss the mechanisms of action by which gut dysbiosis reinforces alcohol use behavior and liver inflammation and injury.

Article Synopsis
  • The study investigates how intrinsic tumor-cell mechanisms, specifically kinases, allow glioblastoma (GBM) cells to evade detection by CD8 T cells, highlighting checkpoint kinase 2 (Chek2) as a key player.
  • An in vivo CRISPR screen identified Chek2 as crucial for tumor cell escape, and its inhibition, along with PD-1 or PD-L1 blockade, improves survival in preclinical glioma models.
  • The research shows that reducing Chek2 enhances antigen presentation and T-cell activation while analysis of human GBMs shows an inverse relationship between Chek2 expression and immune responsiveness, suggesting it as a potential target for improving immune checkpoint therapies in GBM.

Introduction: Crohn's disease (CD) is a major subtype of inflammatory bowel disease (IBD), a spectrum of chronic intestinal disorders caused by dysregulated immune responses to gut microbiota. Although transcriptional and functional changes in a number of immune cell types have been implicated in the pathogenesis of IBD, the cellular interactions and signals that drive these changes have been less well-studied.

Methods: We performed Cellular Indexing of Transcriptomes and Epitopes by sequencing on peripheral blood, colon, and ileal immune cells derived from healthy subjects and patients with CD.
