Multidisciplinary pulmonary embolism response teams (PERTs) have shown that timely triage expedites treatment. The use of artificial intelligence (AI) may help improve pulmonary embolism (PE) management with early CT pulmonary angiogram (CTPA) screening and accelerate PERT coordination. This study aimed to test the clinical validity of an FDA-approved PE AI algorithm.

Article Synopsis
  • Pre-proenkephalin 1 (Penk1) is a pro-neuropeptide linked to analgesic properties and was found to be significantly downregulated in osteoblasts under microgravity conditions, as well as in bone from various models of mechanical unloading.
  • Despite high expression levels in mouse bone, global deletion of Penk1 did not produce noticeable changes in bone metabolism, indicating that its absence does not drastically alter bone structure.
  • In vitro studies showed that inhibiting Penk1 in mature osteoblasts led to changes in gene expression related to bone metabolism, and primary osteoblasts from Penk1 knockout mice exhibited reduced differentiation and metabolic activity, demonstrating Penk1's role in bone response to mechanical changes

Autophagosomes are double-membraned vesicles that engulf cytoplasmic contents, which are ultimately degraded after autophagosome-lysosome fusion. The prevailing view, largely inferred from EM-based studies, was that mammalian autophagosomes evolved from disc-shaped precursors that invaginated and then were closed at the single opening. Many site(s) of origin of these precursors have been proposed.

The sequence of morphological intermediates that leads to mammalian autophagosome formation and closure is a crucial yet poorly understood issue. Previous studies have shown that yeast autophagosomes evolve from cup-shaped phagophores with only one closure point, and mammalian studies have inferred that mammalian phagophores also have single openings. Our superresolution microscopy studies in different human cell lines in conditions of basal and nutrient-deprivation-induced autophagy identified autophagosome precursors with multifocal origins that evolved into unexpected finger-like phagophores with multiple openings before becoming more spherical structures.

Background: Individuals with different mental disorders tend to experience higher rates of colorectal cancer (CRC)-related mortality compared to the general population. Discrepancies in CRC screening behaviors have been suggested as a potential contributing factor to this difference in mortality. However, existing evidence on this topic has been inconclusive and conflicting.
