Neurobehavioral plasticity in the rodent gustatory system induced by regular consumption of a low-calorie sweetener during adolescence.

Habitual consumption of low-calorie sweeteners (LCS) during juvenile-adolescence can lead to greater sugar intake later in life. Here, we investigated if exposure to the LCS Acesulfame Potassium (Ace-K) during this critical period of development reprograms the taste system in a way that would alter hedonic responding for common dietary compounds. Results revealed that early-life LCS intake not only enhanced the avidity for a caloric sugar (fructose) when rats were in a state of caloric need, it increased acceptance of a bitterant (quinine) in Ace-K-exposed rats tested when middle-aged.

Implementation of micelle-enabled C(sp)-C(sp) cross-electrophile coupling in pharmaceutical synthesis.

A sustainable C(sp)-C(sp) cross-electrophile coupling was developed between readily available 5-bromophthalide and 1-benzyl-4-iodopiperidine under micellar conditions, leading to a key intermediate of one of our development compounds. Copper was found to play a crucial role as a co-catalyst in this dual catalysis system. The chemistry and process were successfully demonstrated in a kilo scale to deliver sufficient drug substance to the clinical campaigns.

Oromotor and somatic taste reactivity during sucrose meals reveals internal state and stimulus palatability after gastric bypass in rats.

After Roux-en-Y gastric bypass (RYGB), rats consume less high-energy foods and fluids, though whether this reflects a concomitant change in palatability remains unclear. By measuring behavior during intraorally delivered liquid meals across days (1 water, 8 sucrose sessions), we showed that RYGB rats (RYGB, = 8/sex) consumed less 1.0 M sucrose than their sham surgery counterparts (SHAM, = 8 males, = 11 females) but displayed similarly high levels of ingestive taste reactivity responses at the start of infusions.

Taste- and flavor-guided behaviors following Roux-en-Y gastric bypass in rodent models.

Roux-en-Y gastric bypass (RYGB) surgery is one of the most efficacious treatments for obesity, but the behavioral and physiological mechanisms through which it enacts its effects are not completely understood. The weight loss that follows RYGB surgery is due to some extent to decreased caloric intake. The perception of flavor and the sense of taste undoubtedly contribute to ingestion, and changes in taste sensation and flavor perception may, even in part, propel the altered feeding seen after RYGB surgery.
