Palbociclib, an oral CDK 4/6 inhibitor, was evaluated in a Pediatric Brain Tumor Consortium (PBTC) phase 1 (NCT02255461; PBTC-042) study to treat children and young adults with recurrent, progressive, or refractory brain tumors. The objectives of this study were to characterize the palbociclib population pharmacokinetics in children enrolled on PBTC-042, to conduct a population pharmacodynamic analysis in this patient population, and to perform a simulation study to assess the role of palbociclib exposure on neutropenia and thrombocytopenia. The palbociclib population pharmacokinetics and pharmacodynamics were characterized in this patient population (n = 34 patients; 4.

Background: Cryptosporidiosis is a diarrheal disease that commonly affects calves under 6 weeks old. The causative agent, Cryptosporidium parvum, has been associated with the abundance of specific taxa in the faecal microbiome during active infection. However, the long-term impact of these microbiome shifts, and potential effects on calf growth and health have not yet been explored in depth.

Article Synopsis
  • This study reviews predictors of intra-abdominal injury in pediatric patients with blunt abdominal trauma using data from a Level II trauma center from 2018 to 2022.
  • It found that while various clinical signs and laboratory results were present, a significant number of patients with intra-abdominal injuries (IAI) did not show abnormal lab results, highlighting the complexity in diagnosis.
  • Key independent predictors for IAI included abdominal tenderness, abnormal plain x-rays, and positive ultrasound, suggesting that physical exams and imaging are critical in decision-making for CT scans, rather than relying solely on lab results.

Article Synopsis
  • Large-scale studies are essential to investigate the gut microbiota's complex relationships with health and disease, and tissue preservation methods need evaluation for feasibility in such research.
  • Biopsies from 20 adults with inflammatory bowel disease were preserved using three methods: flash freezing, nucleic acid preservative reagents, and formalin fixation with paraffin embedding (FFPE), with microbiota analyzed using sequencing.
  • Results showed that while preservative reagents can serve as viable alternatives to flash freezing, FFPE samples exhibited distinct community structures, highlighting the potential utility of historical samples despite differences in microbial profiles.
