Article Synopsis
  • Hormone receptor-positive breast cancer shows limited response to immune checkpoint inhibitors (ICIs), but radiation therapy (RT) might enhance the effectiveness of ICIs by boosting immune responses.
  • The study explored the combination of hypofractionated RT with ICIs in a mouse model to see if targeting the primary tumor could delay the emergence of new tumors.
  • Results revealed that while focal RT could control primary tumor growth effectively, adding ICIs did not significantly improve overall survival, suggesting that controlling the primary tumor does not necessarily prevent the development of new cancer lesions.

Introduction: The objective of this study was to examine the interplay of polygenic risk and individual lifestyle factors (and a composite score of lifestyle) as antecedents of CHD in a large multiethnic cohort.

Methods: We used Genetic Epidemiology Resource in Adult Health and Aging (GERA) cohort participants free of CHD at baseline (n = 60,568; 67 % female; 18 % non-European). The individual and joint associations of smoking, Mediterranean diet pattern, level of physical activity and polygenic risk with incident CHD were assessed using Cox regression adjusting for genetic ancestry and non-mediating risk factors.

Purpose: Women with breast cancer (BC) are at high risk of developing cardiovascular disease (CVD). We examined adherence to CVD medications and their association with major CVD events over 14 years of follow-up in the Pathways Heart Study, a prospective study of 4,776 stage I-III BC patients diagnosed from 2005-2013.

Methods: Eligibility included being alive 6 months post-BC diagnosis, with dyslipidemia, hypertension, or diabetes at diagnosis along with ≥1 prior outpatient order or dispensing for a statin, anti-hypertensive, or diabetes medication, respectively, in the 30 months prior.

Article Synopsis
  • Statin drugs, which help lower cholesterol and prevent cardiovascular disease, may cause adverse effects in women, especially new-onset diabetes and muscle weakness.
  • Research in female mice shows that these negative effects are linked to lower levels of the omega-3 fatty acid DHA, as well as issues with cellular function and energy production.
  • Administering fish oil to provide DHA can mitigate these adverse effects, and the study suggests that genetic factors related to the X chromosome may increase women's risk, highlighting the potential of DHA supplementation as a helpful therapy.

Mastocytosis is a heterogeneous disorder characterized by abnormal mast cell accumulation, in which the clinical severity may be explained by distinct molecular mechanisms. This study aimed to explore plasma protein biomarkers associated with systemic mastocytosis subtypes, as well as the cellular origin of the identified proteins. Plasma samples from patients with mastocytosis, including cutaneous mastocytosis (CM), indolent systemic mastocytosis (ISM), and advanced systemic mastocytosis (AdvSM), and a reference group of patients with polycythemia vera, were analyzed by Proximity Extension Assay technology targeting 275 proteins.
