The one-carbon metabolism enzyme bifunctional methylenetetrahydrofolate dehydrogenase/cyclohydrolase 2 (MTHFD2) is among the most overexpressed proteins across tumors and is widely recognized as a promising anticancer target. While MTHFD2 is mainly described as a mitochondrial protein, a new nuclear function is emerging. Here, we observe that nuclear MTHFD2 protein levels and association with chromatin increase following ionizing radiation (IR) in an ataxia telangiectasia mutated (ATM)- and DNA-dependent protein kinase (DNA-PK)-dependent manner.

Nuclear proliferation marker MIB-1 (Ki-67) immunohistochemistry (IHC) is used to examine tumor cell proliferation. However, the diagnostic or prognostic value of the Ki-67 nuclear staining intensity and location, defined as nuclear gradient (NG), has not been assessed. This study examined the potential association between Ki-67 NG and cell cycle phases and its effect on the prognosis of pulmonary typical carcinoid (PTC) tumors.

This paper proposes a two-level, data-driven, digital twin concept for the autonomous landing of aircraft, under some assumptions. It features a digital twin instance (DTI) for model predictive control (MPC); and an innovative, real-time, digital twin prototype for fluid-structure interaction and flight dynamics to inform it. The latter digital twin is based on the linearization about a pre-designed glideslope trajectory of a high-fidelity, viscous, nonlinear computational model for flight dynamics; and its projection onto a low-dimensional approximation subspace to achieve real-time performance, while maintaining accuracy.
