Unlabelled: Newborn screening for cystic fibrosis (CF-NBS) using an IRT-DNA algorithm with a 12 CFTR-variant panel and an IRT/IRT failsafe was officially implemented in the French-speaking Community of Belgium in January 2020. This screening protocol was evaluated after 4 years according to the criteria defined by the European Cystic Fibrosis Society's working group on neonatal screening. Immunoreactive trypsinogen concentration (IRT) was measured on dried blood spots collected between the second and the fourth day of life.

Background: Despite scarce data, invasive mechanical ventilation (MV) is widely suggested as first-line ventilatory support in cardiogenic shock (CS) patients. We assessed the real-life use of different ventilation strategies in CS and their influence on short and mid-term prognosis.

Methods: FRENSHOCK was a prospective registry including 772 CS patients from 49 centers in France.

Article Synopsis
  • The overactivity of the ryanodine receptor type 2 (RyR2) is linked to heart issues like arrhythmias and sudden cardiac death due to improper handling of calcium levels during diastole.
  • Researchers developed a new compound, GM1869, that can both reduce calcium release from RyR2 and enhance calcium uptake through SERCA2a, aiming to improve heart function.
  • GM1869 shows promise over existing drugs like Dantrolene, as it not only lowers calcium spark frequency but also speeds up the decay of calcium transients in cardiomyocytes, potentially aiding individuals prone to arrhythmias.

To discover new multifunctional agents for the treatment of cardiovascular diseases, we designed and synthesized a series of compounds with a cyclopropyl alcohol moiety and evaluated them in biochemical assays. Biological screening identified derivatives with dual activity: preventing Ca leak through ryanodine receptor 2 (RyR2) and enhancing cardiac sarco-endoplasmic reticulum (SR) Ca load by activation of Ca-dependent ATPase 2a (SERCA2a). The compounds that stabilize RyR2 at micro- and nanomolar concentrations are either structurally related to RyR-stabilizing drugs or Rycals or have structures similar to them.

Article Synopsis
  • Cardiogenic shock is a rare condition in adults with congenital heart disease (ACHD), making up only 0.9% of cases in a nationwide registry study in France.
  • The study found that ACHD patients were younger and had fewer risk factors for cardiovascular issues compared to non-ACHD patients, but they still experienced a high rate of severe outcomes after one year, similar to non-ACHD patients.
  • Management strategies differed significantly, with ACHD patients more likely to receive catheterizations and device implantations while less likely to use temporary circulatory support or invasive ventilation.
