Purpose: The study of cell-free DNA (cfDNA) enables sequential analysis of tumor cell-specific genetic alterations in patients with neuroblastoma.

Experimental Design: Eighteen patients with relapsing neuroblastoma having received lorlatinib, a third-generation ALK inhibitor, were identified (SACHA national registry and/or in the institution). cfDNA was analyzed at relapse for nine patients and sequentially for five patients (blood/bone marrow plasma) by performing whole-genome sequencing library construction followed by ALK-targeted ddPCR of the hotspot mutations [F1174L, R1275Q, and I1170N; variant allele fraction (VAF) detection limit 0.

Article Synopsis
  • The study explores the use of stable barium (Ba) sources as substitutes for radioactive iodine (I) in nuclear medicine imaging, aiming to address uncertainties in calibration processes traditionally reliant on liquid radionuclide solutions.* -
  • Researchers created and tested multiple traceable Ba sources within various configurations, conducting quantitative SPECT/CT imaging across several member institutions to compare the effectiveness of Ba versus I.* -
  • Results indicate that, despite initial differences in calibration performance, a specific cross-calibration method allows Ba sources to effectively serve as surrogates for I, showcasing their potential in improving imaging accuracy and efficiency.*

Article Synopsis
  • Atomic and nuclear data are crucial for accurate measurements in liquid scintillation, particularly for β-spectrum calculations.
  • This study focuses on standardizing the activity of two β-emitting radionuclides (Co and Ru/Rh) using 4πβ-γ coincidence and TDCR methods.
  • The results indicate that including atomic effects in the β-spectra improves agreement between the two measurement techniques, and a new stochastic model using Geant4 simulations for TDCR calculations is introduced.

The Bureau International des Poids et Mesures (BIPM) is developing a new transfer instrument to extend its centralized services for assessing the international equivalence of radioactive standards to new radionuclides. A liquid scintillation counter using the triple/double coincidence ratio method is being studied and tested in the CCRI(II)-P1.Co-60 pilot study.

A bilateral comparison to determine the activity concentration of the same I solution was organized. As electron-capture radionuclide with a rather high atomic number, I must be regarded as difficult to measure. The situation is partly exacerbated by the fact that some established standardization methods, like photon-photon coincidence counting, can no longer be applied due to the unavailability of appropriate equipment and expertise.
