Tumor necrosis factor alpha (TNFα), as a key pro-inflammatory cytokine, plays a central role in joint diseases. In recent years, numerous models of TNFα-induced cartilage inflammation have been developed. However, due to the significant differences between these models and the lack of consensus in their construction, it becomes difficult to compare the results of different studies.

Atopic diseases such as Eosinophilic Esophagitis (EoE) often progress into fibrosis (FS-EoE), compromising organ function with limited targeted treatment options. Mechanistic understanding of FS-EoE progression is confounded by the lack of preclinical models and the heavy focus of research on eosinophils themselves. We found that macrophage accumulation precedes esophageal fibrosis in FS-EoE patients.

Importance: A subgroup analysis of a randomized clinical trial established the efficacy of selexipag plus background therapy (monotherapy or double oral therapy [DOT]) vs placebo plus background therapy and found that the addition of selexipag within 6 months had an added benefit. However, the timing of selexipag addition to DOT and the incremental benefit in clinical practice is not well studied.

Objective: To compare triple oral therapy (TOT) consisting of selexipag, endothelin receptor antagonist (ERA), and phosphodiesterase type 5 inhibitor (PDE5i) vs DOT consisting of ERA and PDE5i.

Article Synopsis
  • The study investigates the role of TNFα in inflammatory models for osteoarthritis, focusing on how its effects vary with concentration and chondrocyte growth stages.
  • Bovine chondrocytes were used, cultured in a 3D pellet, and exposed to TNFα for 48 hours, revealing that a concentration of 10 ng/mL was required to significantly induce inflammation.
  • Results showed that higher passages of chondrocytes (especially passage 3) were more sensitive to TNFα, affecting both gene expression of catabolic/inflammatory markers and protein levels of certain cytokines.

Importance: Adaptive expertise helps physicians apply their skills to novel clinical cases and reduce preventable errors. Error management training (EMT) has been shown to improve adaptive expertise with procedural skills; however, its application to cognitive skills in medical education is unclear.

Objective: To evaluate whether EMT improves adaptive expertise when learning the cognitive skill of head computed tomography (CT) interpretation.

Introduction: prostate cancer recurrence after definitive therapy for organ-confined disease often manifests as rising prostate-specific antigen (PSA) levels without clinically overt disease. Gallium prostate-specific membrane antigen, positron emission tomography/computed tomography (GaPSMA PET/CT) imaging plays a major role in the management of recurrent prostate cancer. The purpose of this study was to assess the positivity rate of Ga PSMA PET/CT scans in cases of prostate cancer recurrence, and to compare the results with existing international literature.

Introduction: The severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) pandemic had a significant impact on tuberculosis (TB) control globally, with the number of new TB diagnoses decreasing. Coinfection with some viruses, especially measles, could aggravate TB in children. This is presumably a result of depressed cellular immunity.

Article Synopsis
  • Management of cystic fibrosis (CF) relies on understanding the patient's microbiological status, particularly through evaluating anti-Pseudomonas aeruginosa antibodies, but diversity in bacterial antigens complicates measurement of infection severity.
  • Researchers developed an ELISA test to specifically measure IgG antibody levels against P. aeruginosa porin OprF, allowing for better monitoring of infection status in CF patients.
  • Analysis of serum samples from 310 CF patients revealed that those with chronic colonization had significantly higher OprF antibody levels compared to those with intermittent colonization, highlighting the test's utility in tracking infection progression and treatment effectiveness.

We present a multi-channel delay sampling method to extend imaging depth in high-speed swept-source optical coherence tomography (SS-OCT). A balanced detector captures interference signals, converting them into electrical signals, which are then split into N channels, each with fixed time delays determined by the length of electrical cables. Then, they are digitized by an N-channel acquisition card.

Mitochondrial dysfunction has been linked to both idiopathic and familial forms of Parkinson's disease (PD). We have previously identified RCC1-like (RCC1L) as a protein of the inner mitochondrial membrane important to mitochondrial fusion. Herein, to test whether deficits in RCC1L mitochondrial function might be involved in PD pathology, we have selectively ablated the gene in the dopaminergic (DA) neurons of mice.

Article Synopsis
  • The study focuses on comparing two cartilage treatment methods, autologous chondrocyte transplantation (ACT) and minced cartilage implantation (MCI), specifically their response to inflammation induced by TNFα.
  • Results showed that passaged chondrocytes were more affected by TNFα, leading to negative changes in gene expression and inflammation markers, whereas MCI exhibited lower susceptibility to these inflammatory effects.
  • The findings suggest that MCI may be a more effective approach for cartilage regeneration in osteoarthritis than ACT, but further research is needed to confirm these results for clinical use.
