In experiments on Wistar rats processes nitric oxide production on concentration of anions (NO2-, NO3-), carbamide and polyamines contents were investigated in processes of rats adaptation to acute hypoxia (7% O2 in N2, 30 min) and intermittent hypoxia training (10% O2 in N2, 15 min, 5 cycles daily) during 14 days. NO production by oxygen-dependent and oxygen-independent metabolites paths has been investigated. It is concluded that the disturbances in nitric oxide system induced by acute hypoxia by L-arginine injections may result in acute hypoxia.

Both iNOS expression determined by immunoblotting, and its activity shown by a modified Griess method have been found to predominate in the right ventricle over the left one, as compared to cNOS activity which prevailed in the left ventricle. The latter significantly diminished after an ischemic injury in the left ventricle.

View Article and Find Full Text PDFThe reciprocal changes of NOS and arginase activity during acute myocardial ischaemia (90 min) and reperfusion (180 min) was shown in experiments on chest-closed dogs with spontaneous breathing. NOS activity in the ischemia injured myocardial decreased on 60% while arginase activity increased on 487%. Levels of both alternative pathways of L-arginine metabolism altered reciprocally too.

View Article and Find Full Text PDFThe object of the study was to reveal the existence of receptor interactions of C27-steroid hormone ecdysterone with the surface of various cells (liver and spleen macrophages, thymus and spleen lymphocytes, erythrocytes and hepatocytes from rat organs) in experiments in vitro using ecdysterone immobilized on the nanodispersed iron surface (which did not penetrate the cell during observation) and the model of fast (during 15 s) activation of phospholipid signal system with ecdysterone. The parameters, which are characteristic of cell phospholipid signal system activation, were evaluated (the concentrations of free arachidonic acid, diene conjugates, tromboxane B2 and leukotriene C4) and its alteration in response to free and immobilized ecdysterone introduction in suspensins of intact cells and cells with the surface modified in vivo by pre-injection of antigen (human erythrocytes). Concurrent binding of immbilized ecdysterone with intact rat cells in the presence of free ecdysterone was also investigated.
View Article and Find Full Text PDFThe dynamics of changes in the level of cyclic purine nucleotides was studied in the tissues of rats with experimental D-hypovitaminosis in the range of 5 min-24 h after administration of hormonal doses of polyhydroxylated ecdysterone hydroxysterolne and predecessor of 1.25-dihydroxycholecalciferol (calcitriol)--vitamin D3. The early sharp increase of intracellular pools cEMP in the liver tissue, small intestine, brain and heart mucosa was observed.
