In a previous study, a formulation of methotrexate (MTX) incorporated in the lipid bilayer of 100-nm liposomes in the form of diglyceride ester (MTX-DG, lipophilic prodrug) was developed. In this study, first, the interactions of MTX-DG liposomes with various human and mouse tumor cell lines were studied using fluorescence techniques. The liposomes composed of egg phosphatidylcholine (PC)/yeast phosphatidylinositol/MTX-DG, 8:1:1 by mol, were labeled with fluorescent analogs of PC and MTX-DG.

It was shown the alterations in hydrogen sulfide (H(2)S) metabolism and the development of oxidative and nitrozative stress in cardiovascular system by aging. The administration of pyridoxal-5-phosphate as cofactor of H(2)S synthesizing enzymes restored endogenous H(2)S level and redox state in the heart and aorta tissues. Under these conditions, the following indicators of oxidative stress were significantly decreased in heart and aorta tissues: superoxide generation rate (·0(2)(-)) and hydroxyl (·OH) anion radicals, compared with significantly elevated levels of these parameters in old animals.

As a result of the first gas attack by chlorine undertaken by the German army against the Russian armies on May, 31st, 1915, heavy poisonings have received 9100 people, 6000 of them died. Chemical attack of Germany against Russia was limited by the use chemical gases of suffocating action: chlorine, bromine,phosgene and diphosgene.

MITOCHONDRIAL DYSFUNCTION IN THE AGING HEART IS ACCOMPANIED BY CONSTITUTIVE NO-SYNTHASES UNCOUPLING ON THE BACKGROUND OF OXIDATIVE AND NITROSATIVE STRESS.

We investigated the sensitivity of mitochondrial permeability transition pore (MPTP) opening to its inductor calcium, as well as indicators of oxidative and nitrosative stress in adult and old rat heart mitochondria and heart tissues. The coupling index of constitutive Ca2+/calmodulin-dependent NO-synthase (сNOS) was calculated based on experimentally found parameters. The aging is characterized by oxidative and nitrosative stress, which accompanied by a decrease of the cNOS coupling index and an increased sensitivity of MPTP to calcium.

[Effect of hydrogen sulfide donor NaHs on the functional state of the respiratory chain of the rat heart mitochondria].

In experiments on mitochondria isolated from the heart tissue of adult rats we studied the effects of a donor of hydrogen sulfide, NaHS, on the respiratory chain of the organelles. We found that NaHS (10(-9)-10(-6) mol/l) caused a dose-dependent decrease in the rate of oxygen consumption in the presence of succinate and ADP (state 3 to Chance), and in the absence of ADP (state 4). The decrease in the rate of oxygen consumption in a concentration NaHS 10(-9) mol/l and 10(-8) mol/l associated with an increased conjugation of oxidation and phosphorylation, as evidenced by the increase in the respiratory control, the efficiency of oxidative phosphorylation (ADP/O) is not changed.
