Cellular neurothekeoma (CN) is a benign dermal neoplasm that most often affects the head and neck region and rarely occurs in the oral mucosa. We report a rare case of CN with atypical features on the floor of the mouth and summarize the reported cases of oral CN in English-language literature. A 62-year-old woman presented with a 6-month history of a painless mass on the floor of the mouth.

Aims: Radiation caries (RC) is a highly prevalent and chronic complication of head and neck radiotherapy (HNRT) and presents a challenge for clinicians and patients. The present study aimed to assess the impact of RC on the morbidity and mortality outcomes of head and neck squamous cell carcinoma (HNSCC) patients.

Methods And Results: Patients were divided into three groups: (1) RC (n = 20), (2) control (n = 20), and (3) edentulous (n = 20).

Oral cancer refers to malignant tumors, of which 90% are squamous cell carcinomas (OSCCs). These malignancies exhibit rapid progression, poor prognosis, and often mutilating therapeutical approaches. The determination of a prophylactic and/or therapeutic antitumor role of the polyphenolic extract would be relevant in developing new tools for prevention and treatment.

Background: Fibroblast growth factor receptor 1 is a potential prognostic factor for tongue squamous cell carcinoma and is associated with oral epithelial dysplasia grade in oral leukoplakia.

Methods: Thirty cases of tongue squamous cell carcinoma and 30 cases of oral leukoplakia were analyzed. Fibroblast growth factor receptor 1 and phosphorylated Akt protein expression were analyzed by immunohistochemistry and quantified using a digital algorithm.

Background: Rhabdomyosarcoma (RMS) harboring EWSR1/FUS-TFCP2 fusions has been recently described as a distinct form of RMS with an aggressive course and predilection for the craniofacial bones, especially the jaws.

Methods: We report three new cases of this rare entity, two from Brazil and one from Guatemala, with detailed clinicopathologic, immunohistochemical, and molecular descriptions. Additionally, we explored the English-language literature searching RMS with TFCP2 rearrangement or typical immunophenotype with co-expression of AE1/AE3 and ALK in the head and neck region.

Aspergillosis is a fungal infection caused by species, which is contracted through spores that colonize the respiratory tract, causing rhinosinusitis and pulmonary infections. Oral aspergillosis is rare and, when present, may cause soft tissue and bone destruction, generally in immunodeficient patients. Mandibular osteomyelitis is even rarer, with few cases reported in the literature.

Hybrid peripheral nerve sheath tumors show combined features of more than one type of conventional benign peripheral nerve sheath tumors. There are few cases reported of hybrid peripheral nerve sheath tumors in the head and neck region. A 68-year-old female patient was referred for evaluation of an oral swelling lasting five years.

Article Synopsis
  • Spindle cell carcinoma (SpCC) is a rare form of squamous cell carcinoma that features both carcinomatous and sarcomatous traits, making diagnosis challenging.
  • A case study highlighted a 50-year-old woman previously treated for melanoma, who presented with a painful lip lesion diagnosed as SpCC after surgical analysis showed specific immunohistochemical markers.
  • The patient experienced metastases within a year, leading to her death after three years, emphasizing the need for detailed microscopic and immunohistochemical evaluations for accurate diagnosis.

Objective: To analyze the proteomic profile of salivary pleomorphic adenoma (PA) and carcinoma ex pleomorphic adenoma (CXPA) samples and correlate them with the malignant transformation of the PA.

Materials And Methods: Thirty samples (10 PA, 16 CXPA, and 4 residual PA) were microdissected and submitted to liquid chromatography-tandem mass spectrometry (LC-MS/MS). The proteomic data and protein identification were analyzed through LC-MS/MS spectra using the MaxQuant software.

A proper antibody panel selection is one of the most important factors to reach an adequate diagnosis in challenging cases. This retrospective study was designed to determine the contribution of immunohistochemistry (IHC) in the primary diagnosis of oral diseases in one of the main services of oral pathology in the State of São Paulo, Brazil, and to identify the most common antibodies used, and recommend diagnostic algorithms based on our experience with challenging lesions. A total of 1698 IHC stains were performed in 401 cases from a total of 28,804 cases received from public dental clinics and private dental practitioners within a period of 13 years, representing a frequency of 1.

Article Synopsis
  • - The study focused on Burkitt lymphoma affecting the head and neck (BLHN) in pediatric patients from Africa, Guatemala, and Brazil, revealing common clinical and anatomical characteristics across these regions.
  • - Among the 52 cases reviewed, a significant majority (82.69%) were children, with common symptoms including tumors, cervical lymphadenopathy, and bone destruction, and typical features associated with Burkitt lymphoma were observed.
  • - Immunohistochemical analysis showed uniform expression of specific markers like CD20 and high Ki-67 indices, with Epstein-Barr virus (EBV) present in about 62% of cases, indicating similarities in the disease profile across the studied populations.

Oral tongue squamous cell carcinoma (OTSCC) causes over 350,000 cases annually and particularly impacts populations in developing countries. Smoking and alcohol consumption are major risk factors. Determining the role of the tumor immune microenvironment (TIME) in OTSCC outcomes can elucidate immune mechanisms behind disease progression, and can potentially identify prognostic biomarkers.

The role of digital pathology in remote reporting has seen an increase during the COVID-19 pandemic. Recently, recommendations had been made regarding the urgent need of reorganizing head and neck cancer diagnostic services to provide a safe work environment for the staff. A total of 162 glass slides from 109 patients over a period of 5 weeks were included in this validation and were assessed by all pathologists in both analyses (digital and conventional) to allow intraobserver comparison.

Article Synopsis
  • Neuroblastoma is the most common solid cancer found outside the brain in infants, often originating in the adrenal gland, but it rarely spreads to the head and neck area, as seen in two reported cases where mandibular swelling indicated advanced disease.
  • The first case involves a 4-year-old boy with painful swelling in the left jaw and destructive lesions confirmed through imaging, while the second case features a 3-year-old boy with swelling in the right jaw that also showed significant bone destruction.
  • Both cases displayed aggressive tumor characteristics through microscopic analysis and high proliferation rates, leading to treatment involving chemotherapy and surgery to manage the cancer and its spread to multiple skeletal sites.

Article Synopsis
  • - Osteopontin (OPN) is a protein that interacts with calcium and is crucial for various bodily functions, particularly in bone cell activities and mineralization, but it also plays a role in diseases like cancer and atherosclerosis.
  • - Research indicates that OPN may contribute to the development of oral cancer by promoting aggressive cell behaviors through the PI3K/AKT/mTOR signaling pathway, which can enhance cell growth, invasion, and resistance to treatment.
  • - The text reviews the mechanisms by which OPN is involved in oral cancer and explores its potential use in clinical settings for diagnosis and prognosis of the disease.
