The amino-termina, noncatalytic half of Src contains two domains, designated the Src homology 2 (SH2) and Src homology 3 (SH3) domains, that are highly conserved among members of the Src family of tyrosine kinases. The SH2 domain (which can be further divided into the B and C homology boxes) and the SH3 domain (also referred to as the A box) are also found in several proteins otherwise unrelated to protein tyrosine kinases. It is believed that these domains are important for directing specific protein-protein interactions necessary for the proper functioning of Src.

We report here that stimulation of platelets by cross-linking antibodies to Fc gamma RII or by treatment with an anti-CD9 monoclonal antibody, which acts through Fc gamma RII, causes an induction of tyrosine phosphorylation of multiple platelet proteins. Although the profile of tyrosine-phosphorylated proteins induced by stimulation of this Fc receptor was similar to that induced by thrombin, an additional 40-kDa phosphorylated protein was also detected.

The intracellular signalling pathways that mediate changes in cell behavior induced by extracellular matrix and cell adhesion molecules are poorly understood. Studies on the regulation of tyrosine phosphorylation in platelets indicate that cell-to-cell aggregation mediated by fibrinogen binding to its integrin-family receptor, GP IIb-IIIa, and events regulated by the putative adhesion receptor, GP IV (CD36), involve tyrosine phosphorylation. Structural information on the LIF receptor is not yet available, whereas recent cloning of the IL-6 receptor has shown it to be bipartite, with a signal-transducing subunit that lacks sequence homology to known protein kinases and produces second messengers of unknown nature. The molecular nature of the mechanisms which LIF and IL-6 use to induce cell differentiation is not known.

Growth factors and transforming proteins that activate tyrosine phosphorylation have been shown to cause an increased labeling of 3-phosphate-containing phosphatidylinositols. Turnover correlates with the formation of a complex between phosphatidylinositol 3-kinase, the activated protein-tyrosine kinase, and other proteins thought to participate in transmembrane signaling. To further examine the functional importance of phosphorylation at Tyr416, we have eliminated this phosphoacceptor site in four functionally unique mutant forms of the c-src gene product that are phosphorylated on Tyr416 in vivo. High-resolution SDS-PAGE allowed analysis of both the 'fibroblast' 60-kDa form and a variant, 61-kDa neuron-specific form of the c-src gene product which is encoded by an alternately processed c-src mRNA. Studies of microdissected brain regions showed that all CNS regions contained both forms of the enzyme, the 61-kDa form predominating in most regions with high content of gray matter and high density of synapses.
