Development of a novel MATLAB-based framework for implementing mechanical joint stability constraints within OpenSim musculoskeletal models.

The Static Optimization (SO) solver in OpenSim estimates muscle activations and forces that only equilibrate applied moments. In this study, SO was enhanced through an open-access MATLAB interface, where calculated muscle activations can additionally satisfy crucial mechanical stability requirements. This Stability-Constrained SO (SCSO) is applicable to many OpenSim models and can potentially produce more biofidelic results than SO alone, especially when antagonistic muscle co-contraction is required to stabilize body joints.

DFMO and 5-Azacytidine Increase M1 Macrophages in the Tumor Microenvironment of Murine Ovarian Cancer.

Effects of foot position on skin structural deformation.

As the largest and most superficial organ, the skin is well positioned for receiving sensory information from the environment. It is conceivable that changes in posture could result in deformations of the skin and subsequent changes in skin material properties. Specifically, the ankle and metatarsophalangeal joints have the capability to undergo large postural alterations with the potential to induce large structural deformations in the skin of the foot.

Maintenance of spine stability is considered to be a critical component of spine health. Ross et al. A Novel Mechanism of High Dose Radiation Sensitization by Metformin.

Metformin, the most widely used treatment for diabetes, is lethal to cancer cells and increases in toxicity when used in combination with radiation. In addition to various molecular and metabolic mechanisms that have been previously proposed, the studies presented provide evidence of an additional, novel mechanism of sensitization following high dose radiotherapy; the magnitude of sensitization depends on the microenvironmental levels of glucose and oxygen which are in turn affected by high dose radiation. Comparison between patient-reported outcomes after enucleation and proton beam radiotherapy for uveal melanomas: a 2-year cohort study.

Background: Uveal melanomas affect 2-8 per million Europeans each year. Approximately 35%, are treated by enucleation. Proton beam radiotherapy (PBR) can be an eye-conserving alternative to enucleation for patients who wish to retain the eye.

Defining UHRF1 Domains that Support Maintenance of Human Colon Cancer DNA Methylation and Oncogenic Properties.

UHRF1 facilitates the establishment and maintenance of DNA methylation patterns in mammalian cells. The establishment domains are defined, including E3 ligase function, but the maintenance domains are poorly characterized. Here, we demonstrate that UHRF1 histone- and hemimethylated DNA binding functions, but not E3 ligase activity, maintain cancer-specific DNA methylation in human colorectal cancer (CRC) cells.

4-Dimensional computed tomography-based ventilation and compliance images for quantification of radiation-induced changes in pulmonary function.

Introduction: 4-Dimensional computed tomography (4DCT)-based ventilation imaging is a promising technique for evaluating pulmonary function, but lung elasticity and mechanics are usually not part of the ventilation image analysis. In this study we demonstrate a 4DCT-based imaging technique that can be used to calculate regional lung compliance changes after radiotherapy (RT).

Objective: Shared decision-making exists to reconcile healthcare practitioners' responsibilities to respect patients' autonomy whilst ensuring well-made decisions. Patients sometimes make unprompted requests for procedures that carry medical and other risks, such as risk-reducing mastectomy (RRM). Faced with pre-formed decisions into which they have had little input, it is unclear how practitioners can reconcile respecting autonomy with ensuring well-made decisions.

Does other-affirmation increase self-directed exposure to and persuasiveness of a threatening anti-alcohol message?

Objective: Self-affirmation of personal values can reduce defensive responses to threatening health promotion messages, probably because it induces a positive and expansive view of the self. However, coping with threat is also an interpersonal process. We developed other-affirmation inductions that focus on values held by others.

ITPase deficiency causes a Martsolf-like syndrome with a lethal infantile dilated cardiomyopathy.

Typical Martsolf syndrome is characterized by congenital cataracts, postnatal microcephaly, developmental delay, hypotonia, short stature and biallelic hypomorphic mutations in either RAB3GAP1 or RAB3GAP2. Genetic analysis of 85 unrelated "mutation negative" probands with Martsolf or Martsolf-like syndromes identified two individuals with different homozygous null mutations in ITPA, the gene encoding inosine triphosphate pyrophosphatase (ITPase). Both probands were from multiplex families with a consistent, lethal and highly distinctive disorder; a Martsolf-like syndrome with infantile-onset dilated cardiomyopathy.

Aging-like Spontaneous Epigenetic Silencing Facilitates Wnt Activation, Stemness, and Braf-Induced Tumorigenesis.

We addressed the precursor role of aging-like spontaneous promoter DNA hypermethylation in initiating tumorigenesis. Using mouse colon-derived organoids, we show that promoter hypermethylation spontaneously arises in cells mimicking the human aging-like phenotype. The silenced genes activate the Wnt pathway, causing a stem-like state and differentiation defects.

Discriminating spatiotemporal movement strategies during spine flexion-extension in healthy individuals.

Background Context: The spine is an anatomically complex system with numerous degrees of freedom. Due to this anatomical complexity, it is likely that multiple motor control options exist to complete a given task.

Determining binding of polycyclic aromatic hydrocarbons to CTABr micelles using semi-equilibrium dialysis techniques.

Micellar enhanced ultrafiltration (MEUF) has been shown to be an effective removal technique for a variety of trace contaminants in wastewater, especially for water soluble low molecular weight organic contaminants. In MEUF, contaminants first partition into surfactant micelles prior to removal by ultrafiltration, where the contaminants are below the molecular weight cut-off of the ultrafiltration membrane but the micelles are above the cut-off. Binding constants of selected Polycyclic aromatic hydrocarbons (PAHs) such as naphthalene, phenanthrene, pyrene and fluorene have been determined in order to examine the removal efficiency of MEUF for PAHs with Cetyltrimethylammonium bromide (CTABr) using a semi equilibrium dialysis method.

Public Health Quality Improvement Exchange: A Tool to Support Advancements in Public Health Practice.

Objectives: The Public Health Quality Improvement Exchange(PHQIX) is a free, openly available online community that supports public health practitioners in the rapidly evolving landscape of public health quality improvement (QI). This article's objective is to describe the user-centered development of PHQIX and its current content and examine how elements of a QI initiative may vary by an organization's characteristics or QI experience.

Methods: PHQIX was developed by taking a user-centered iterative design approach, seeking early and continued input from users to gather requirements for the website.

Fecal indicator bacteria levels at beaches in the Florida Keys after Hurricane Irma.

Hurricanes cause infrastructure failures which can lead to contamination of impacted areas. The objective of the current study was to evaluate whether Hurricane Irma contributed towards sewage contamination of coastal beaches. Through this study we evaluated indicators of fecal pollution (fecal indicator bacteria [FIB], enterococci and fecal coliform) and physico-chemical parameters (salinity, pH, turbidity, and temperature) in coastal waters of the Florida Keys shortly after the hurricane.

Kinetic modeling of tumor regression incorporating the concept of cancer stem-like cells for patients with locally advanced lung cancer.

Background: Personalized medicine for patients receiving radiation therapy remains an elusive goal due, in part, to the limits in our understanding of the underlying mechanisms governing tumor response to radiation. The purpose of this study was to develop a kinetic model, in the context of locally advanced lung cancer, connecting cancer cell subpopulations with tumor volumes measured during the course of radiation treatment for understanding treatment outcome for individual patients.

Methods: The kinetic model consists of three cell compartments: cancer stem-like cells (CSCs), non-stem tumor cells (TCs) and dead cells (DCs).

Comparative toxicity of two diluted bitumens to developing yellow perch (Perca flavescens).

Increasing demand for diluted bitumen (dilbit) has led to the development of the oil sands industry and the expansion of transcontinental pipelines. Dilbit is an unresolved complex mixture with variable diluent and bitumen composition. Thus, it is important to understand the effects of the two most transported dilbits in Canada, Access Western Blend (AWB) and Cold Lake Blend (CLB) on a North America native and freshwater fish species, the yellow perch (Perca flavescens).

Reconciling the theory and reality of shared decision-making: A "matching" approach to practitioner leadership.

Shared decision making (SDM) evolved to resolve tension between patients' entitlement to make health-care decisions and practitioners' responsibility to protect patients' interests. Implicitly assuming that patients are willing and able to make "good" decisions, SDM proponents suggest that patients and practitioners negotiate decisions. In practice, patients often do not wish to participate in decisions, or cannot make good decisions.

Increased Substance P Immunoreactivity in Ipsilateral Knee Cartilage of Rats Exposed to Lumbar Spine Injury.

Objective: The present study aimed to investigate whether experimentally induced lumbar facet-joint OA lead to degenerative changes and enhanced SP expression within the ipsilateral neurosegmentally linked tibiofemoral cartilage.

Methods: Adult male Sprague-Dawley rats were assigned to left side L5-L6 facet mechanical compression injury (surgery) ( = 6), L5-L6 facet exposure with no compression (sham) ( = 5), or naïve (no surgery) ( = 4) groups. The morphology of the tibiofemoral articular cartilage was assessed using a modified Mankin scoring system.

Differential effects of muscle fatigue on dynamic spine stability: Implications for injury risk.

This study was designed to assess the utility of using a measure of dynamic spine stability in an unfatigued, rested state as a predictor of dynamic spine stability in a challenged, fatigued state. Participants completed three trials (Day 1: Rested, Fatigued; Day 2: Recovery) during which the dynamic stability of the spine was assessed over 30 repeated flexion/extension motions using maximum finite-cycle Lyapunov exponents. Multiple sets of dynamic trunk extensions were performed to fatigue the trunk extensor muscles.
