The developmental origins of health and disease theory suggests that environmental exposures during early life, particularly during prenatal life, can greatly influence health status later in life. Irregular light-dark cycles, such as those experienced during shift work, result in the repeated disruption of circadian rhythms, which negatively impacts physiological and behavioral cycles. The purpose of our study was to assess parameters in the developing mouse embryo and fetus using high frequency ultrasound when exposed to circadian disruption.

Array tomography (AT) allows one to localize sub-cellular components within the structural context of cells in 3D through the imaging of serial sections. Using this technique, the z-resolution can be improved physically by cutting ultra-thin sections. Nevertheless, conventional immunofluorescence staining of those sections is time consuming and requires relatively large amounts of costly antibody solutions.

Traditionally, peripheral sensory neurons are assumed as the exclusive transducers of external stimuli. Current research moves epidermal keratinocytes into focus as sensors and transmitters of nociceptive and non-nociceptive sensations, tightly interacting with intraepidermal nerve fibers at the neuro-cutaneous unit. In animal models, epidermal cells establish close contacts and ensheath sensory neurites.

In correlative light and electron microscopy (CLEM), the fluorescent images must be registered to the EM images with high precision. Due to the different contrast of EM and fluorescence images, automated correlation-based alignment is not directly possible, and registration is often done by hand using a fluorescent stain, or semi-automatically with fiducial markers. We introduce "DeepCLEM", a fully automated CLEM registration workflow.

Introduction: While a large proportion of people with HIV (PWH) have experienced SARS-CoV-2 infections, there is uncertainty about the role of HIV disease severity on COVID-19 outcomes, especially in lower-income settings. We studied the association of mortality with characteristics of HIV severity and management, and vaccination, among adult PWH.

Methods: We analysed observational cohort data on all PWH aged ≥15 years experiencing a diagnosed SARS-CoV-2 infection (until March 2022), who accessed public sector healthcare in the Western Cape province of South Africa.

To enable flexible and controlled research on personality, information processing, and interactions in socio-emotional contexts, the availability of highly controlled stimulus material, especially trait words and related attributes, is indispensable. Existing word databases contain mainly nouns and rating dimensions, and their role in studies within socio-emotional contexts are limited. This study aimed to create an English list of traits (ELoT), a database containing 500 trait adjectives rated by a large sample (n = 822, 57.

The Long-term In-situ Test (LIT) of the Colloid Formation and Migration project (CFM) at the Grimsel Test Site, investigates the generation of bentonite colloids and, hence, radionuclide mobilization within a well-defined and controlled shear zone in a crystalline rock. In this context, the determination of radionuclide aqueous speciation is essential to understand whether radionuclides are easily transported or immobilized by precipitation or uptake processes in the bentonite barrier included in a repository concept for nuclear waste, and mimic in the LIT experiment. The objective of this work is to determine the aqueous speciation of seven radionuclides (i.

At the end of the first larval stage, the nematode developing in harsh environmental conditions is able to choose an alternative developmental path called the dauer diapause. Dauer larvae exhibit different physiology and behaviors from non-dauer larvae. Using focused ion beam-scanning electron microscopy (FIB-SEM), we volumetrically reconstructed the anterior sensory apparatus of dauer larvae with unprecedented precision.

The mobility of heavy metal contaminants and radionuclides in the environment is directly controlled by their interactions with charged mineral surfaces, hence an assessment of their potential toxicity, e.g. in the context of radioactive waste disposal sites, requires understanding of sorption processes on the molecular level.

The processing of self-referential information can be influenced by transcranial magnetic stimulation (TMS). The present randomized controlled study investigated whether similar effects can be elicited through the application of transcranial direct current stimulation (tDCS) regarding the "self-serving bias" (SSB) and the "mnemic neglect effect" (MNE). Seventy-five healthy males ( = 25; SD = 4.

People maintain a positive self-concept through positive self-appraisals (Self-Serving Bias Effect, SSBE) and a diminished memory for self-threatening information (Mnemic-Neglect Effect, MNE). Other people also influence a person's self-concept. This study investigated SSBE and MNE in 60 females by using a trait-judgment paradigm applying two perspectives (self- and third-person appraisals) and a recall task.

In this study, we present the Aachen List of Trait Words (ALoT), including a total of 606 German adjectives with English translations, describing personality traits. The lack of ratings regarding the social desirability of traits led us to create a German trait database. Ratings of valence and social desirability were obtained from 100 participants.

Microvascular endothelial cells are an essential part of many biological barriers, such as the blood⁻brain barrier (BBB) and the endothelium of the arteries and veins. A reversible opening strategy to increase the permeability of drugs across the BBB could lead to improved therapies due to enhanced drug bioavailability. Vanilloids, such as capsaicin, are known to reversibly open tight junctions of epithelial and endothelial cells.

Array Tomography (AT) is a relatively easy-to-use and yet powerful method to put molecular identity in its full ultrastructural context. Ultrathin sections are stained with fluorophores and then imaged by light and afterward by electron microscopy to obtain a correlated view of a region of interest: its ultrastructure and specific staining. By combining AT with high-pressure freezing for superior structural preservation and superresolution light microscopy, even small subcellular structures can be mapped in 3D.

Correlating molecular labeling at the ultrastructural level with high confidence remains challenging. Array tomography (AT) allows for a combination of fluorescence and electron microscopy (EM) to visualize subcellular protein localization on serial EM sections. Here, we describe an application for AT that combines near-native tissue preservation via high-pressure freezing and freeze substitution with super-resolution light microscopy and high-resolution scanning electron microscopy (SEM) analysis on the same section.

Absorption of cyanidin-based anthocyanins is not fully understood with respect to dose or anthocyanin structure. In feeding studies using whole foods, nonacylated anthocyanins are more bioavailable than their acylated counterparts, but the extent to which plant matrix determines relative bioavailability of anthocyanins is unknown. Using juice of purple carrots to circumvent matrix effects, a feeding trial was conducted to determine relative bioavailability of acylated and nonacylated anthocyanins and to assess dose-response effects.

We examined the characteristics of ascorbic acid (ASC) level, dehydroascorbate (DHA) level, and the ASC-DHA redox status in the leaflets of two soybean cultivars grown in a field environment and exposed to elevated ozone (O(3)) levels. These two cultivars, one that preliminary evidence indicated to be O(3)-tolerant (cv Essex), and one that was indicated to be O(3)-sensitive (cv Forrest), were grown in open-top chambers during the summer of 1997. The plants were exposed daily to a controlled, moderately high O(3) level ( approximately 58 nl l(-1) air) in the light, beginning at the seedling stage and continuing to bean maturity.
