Article Synopsis
  • DNA double-strand breaks occur due to genotoxic agents and cellular activities, playing a critical role in various physiological processes.
  • In lymphocytes, these DNA breaks trigger a wide-ranging transcriptional program that goes beyond the typical DNA damage response, influencing important genes for lymphocyte development.
  • Both physiological and genotoxic DNA breaks affect the same gene expressions, indicating that genotoxic damage could interfere with normal cellular functions related to lymphocyte development.

Whereas interleukin-10 (IL-10) is an anti-inflammatory cytokine known to regulate macrophage activation, its full mechanism of action remains incompletely defined. In a screen to identify novel IL-10-induced genes, we cloned the mouse ortholog of human ABIN-3 (also termed LIND). ABIN-3 expression was induced selectively by IL-10 in both mouse and human mononuclear phagocytes coordinately undergoing proinflammatory responses.

Striking similarities continue to emerge between the mammalian and Drosophila JAK/STAT signaling pathway. However, until now there has not been the ability to monitor global pathway activity during development. We have generated a transgenic animal with a JAK/STAT responsive reporter gene that can be used to monitor pathway activation in whole Drosophila embryos.
