The results obtained show the essential changes in functional state of hepatocyte's plasmatic membrane due to the implantation of human ApoA1 gene to the rat liver. The changes in phospholipid composition, hyperpolarization, increase in activity of membrane bound enzymes, cytochrome P-450 and biosynthesis of liver total proteins have been found. The essential changes characterizing cell effect were more marked in the adult rats, and membrane effect in the old ones.

In hypercholesterolemia, there was a considerable weakening of a vasodilatory action of atherogenic LP fractions--LDL and VLDL. A vasodilatory effect of HDLP was not changed.

The presence of generalized cell reaction is evidenced from the fact than biosynthesis of summary proteins gets enhanced during the above changes.

The study revealed age-related changes in the content and ratio of apoproteins which are part of high-density lipoproteins (HDL), as well as the development of dysapolipoproteinemia. The most significant fall (by 46.7%) was found in the content of the key HDL apoprotein--apoA1, and rise (by 107%)--in apoE.

The content of cholesterol and cholesterol phospholipids ratio were found to increase with age. The total content of phospholipids remained unchanged during ageing, while their composition varied.

The lipid matrix of membranes was found to undergo appropriate changes with age. The most significant shifts are manifested under functional and metabolic stresses.

Olivomycin, administered simultaneously with long-term cholesterol loading, and streptomycin, injected within 3 days before cholesterol loading, were used as inhibitors. These antibiotics prevented hypercholesterolemia, hyperlipoproteinemia and atherosclerosis development.

Essentiale normalizes the cholesterol metabolism, adebit normalizes the carbohydrate metabolism. Results of the treatment indicate that elderly and old, in particular, showed an improvement of the clinical condition of the disease, normalization of the lipid metabolism.

Changes in pH and pO2 of the blood have been studied for age peculiarities of their effect on the glycolysis rate and the content of ATP and 2,3-diphosphoglycerate (2,3-DPG) in erythrocytes (in vitro). The fresh venous blood of practically healthy young (aged 20-29) and old (aged 75-85) people was used. Acidosis was shown to induce inhibition of glycolysis and decrease of the ATP and 2.

A fall of the hexokinase activity is shown to be one of the reasons of a significant decrease in the glycolysis rate. The total ATPase activity in erythrocytes does not change with the age.

It is shown that the glycolysis rate and the content of ATP decrease under ageing by 16% and 21%, respectively. The concentration of 2,3-DPG lowers insignificantly (by 8%).

It is shown that under conditions of experimental limitation of coronary blood flow in rabbits the intensity of radioactive label (32P) incorporation into creatine phosphate lowers essentially. This evidences for inhibition of creatine phosphorylation in mitochondria under ischemia due to a decrease in the activity of a mitochondrial isoenzyme of creatine kinase.

However, redistribution of cholesterol occurred between various classes of lipoproteins. There was a decrease of cholesterol content in high density lipoproteins (LPHD) and an increase in cholesterol content in lipoproteins of very low and low density.

Content of ATP and ADP and the Na+, K+-ATPase activity were decreased while the level of Pi was increased in rat brain cortex during ageing. Administration of vasopressin caused an increase in lactate, Pi and Na+, K+-ATPase activity, whereas creatine phosphate decreased in the adult rat hypothalamus.
