The influence of different cyclodextrin derivatives on the in vitro permeability of diclofenac sodium through pig cornea was investigated and compared to the commercial product Voltaren ophtha. (Hydroxypropyl)-beta-cyclodextrin (HP beta CD) and two amorphous methylated cyclodextrins with different degrees of substitution were used. Diabetes incidence in BB rats is 60-80% and our aim was to investigate whether it is possible to characterize those rats destined to develop diabetes. While the genetic background as well as the environmental factors affecting BB-rat littermates are very similar, body weight reflects some existing variance. Segregating families of beet (Beta vulgaris) were used to verify the monofactorial inheritance of two enzyme-coding loci, leucine aminopeptidase (Lap1) and glutamate oxaloacetate transaminase (Got3). With a series of primary trisomies and using three methods to discriminate between the critical trisomic (the locus is situated on the triplicated chromosome) and the non-critical ones, it was possible to allocate the two loci to beet chromosomes I and II, respectively. For the locus Lap1 distorted segregation ratios were estimated, and the incorporation of three alleles into one plant was attempted.

This study examined the expression of mRNA coding for the 65-kDa heat-shock protein (HSP) in rat islet cells of different functional states and different ages. In addition, beta cells and non-beta cells purified by fluorescence-activated cell sorting were studied. Total RNA from islet cells and insulin-producing RINm5F cells was isolated and analyzed by Northern blotting using a cDNA probe coding for the human homologue to the mycobacterial 65-kDa HSP, after which blots were quantified by densitometric scanning.

Prophylactic insulin treatment is effective in preventing diabetes in animal models of insulin-dependent diabetes mellitus (IDDM) but the safety of such preventive treatment in prediabetic human subjects remains unclear; insulin is a potential autoantigen that could accelerate beta-cell decompensation and onset of IDDM. Antigen expression in type 1 (insulin-dependent) diabetes may depend on the functional state of beta cells. At birth, beta cells are immature, produce only a basal amount of insulin, and are unresponsive to glucose--but are sensitive to glucagon and arginine. beta cells of spontaneously diabetic BB rats were stimulated for the first 6 days after birth by glucose with glucagon or arginine to accelerate beta cell maturation, and possibly to induce antigen expression and tolerance.

Alien monosomic additions in beet (Beta vulgaris), each carrying one of the nine chromosomes of B. procumbens, were grown in vivo and in vitro to study the effect of the alien chromosomes on plant development. All additional chromosomes caused a reduction of the growth rate in vivo, which, in one case was so strong that some of the plants died as seedlings.

Coagulation parameters, platelet aggregation, and thromboxane production as well as metabolic parameters were measured in 31 diabetic patients, 12 without and 19 with clinically manifest late complications, and in 14 healthy control subjects. Spontaneous in vitro aggregation as well as ADP, collagen, and arachidonic acid induced aggregation were higher in both groups of diabetic patients, without an increase in thromboxane B2 production. In diabetic patients with late complications an increase in fibrinogen, fibrinogen cyanogen bromide peptide, factor VIII related antigen, C1-esterase inhibitor, and antithrombin III was observed in comparison to healthy subjects.

A tetraploid (2n=36) interspecific hybrid was obtained involving three species belonging to three different sections of Beta. The hybrid was highly sterile and did not show apomixis. The activity of three combinations of regular and NPH human insulin (recombinant DNA) has been compared to that of an intermediate-acting pork insulin in a crossover study in type I diabetic subjects. After a prephase, the patients injected each insulin subcutaneously for 1 wk in different order. During the 5-wk period, daily glucose profiles were self-monitored, except at the end of each week, when blood was sampled for the determination of glucose, HbA1, glycosylated albumin, C-peptide, glucagon, and routine laboratory parameters.

A rate-adjusting pacemaker system is described which uses central venous oxygen saturation (SO2) for the regulation of the pacing rate. This system was tested externally in ten patients with chronically implanted VVI units. There was an average increase in cardiac output of 18% with the SO2-regulated pacemaker as compared to the situation during fixed rate stimulation.
