Neuro-oncology biobanks are critical for the implementation of a precision medicine program. In this perspective, we review our first year experience of a brain tumor biobank with integrated next generation sequencing. From our experience, we describe the critical role of the neurosurgeon in diagnosis, research, and precision medicine efforts.

Article Synopsis
  • * The study uses ELIC, a prokaryotic ion channel, to identify key phospholipid interaction sites, revealing structural features that influence these interactions.
  • * Findings show that the flexibility of the M4 helix and its lipid-binding capability are essential for the channel's response to agonists, highlighting the importance of lipid interactions in the function of pLGICs.

Background: Clozapine is a second-generation antipsychotic typically used for refractory schizophrenia or otherwise psychotic pathology. There are no FDA or manufacturer guidelines for use of clozapine in pediatric population. We investigated the current state of research concerning the use of clozapine in pediatric patients.

Severe peripheral nerve injury occasionally requires urgent nerve grafting especially with significant separation of the proximal and distal ends of the injured nerve. Proper reinnervation to provide continued sensory and motor function is essential especially in the pediatric population. These patients would suffer lifelong disability without correction, yet have significantly improved regenerative capacity with prompt and effective management, making nerve grafts an ideal choice for complete nerve transection.

Article Synopsis
  • Continuous-wave electron paramagnetic resonance spectroscopy (CW-EPR) and site-directed spin labeling (SDSL) are effective techniques used to study protein structures and dynamics, particularly in integral membrane proteins.
  • These methods have demonstrated their utility in understanding the structural changes associated with the biological functions of these proteins.
  • The chapter provides practical guidance for performing spin-labeling, reconstituting proteins in liposomes, and conducting CW-EPR measurements specifically for the bacterial potassium channel KcsA.

The Erwinia chrysanthemi ligand-gated ion channel, ELIC, is considered an excellent structural and functional surrogate for the whole pentameric ligand-gated ion channel family. Despite its simplicity, ELIC is structurally capable of undergoing ligand-dependent activation and a concomitant desensitization process. To determine at the molecular level the structural changes underlying ELIC's function, it is desirable to produce large quantities of protein.
