View Article and Find Full Text PDFObjective: The purpose of this study was to apply clustering methods to identify and characterize prediabetes phenotypes and their relationships with treatment arm and type 2 diabetes (T2D) outcomes in the Diabetes Prevention Program (DPP), and to compare the utility of additional clustering measures in phenotype characterization and T2D risk stratification.
Research Design And Methods: This was a secondary analysis of data from a subset of participants (n=994) from the previously completed Diabetes Prevention Program trial. Unsupervised k-means clustering analysis was applied to derive the optimal number of clusters of participants based on common clinical risk factors alone or common risk factors plus more comprehensive measures of glucose tolerance and body composition.
Objectives: Environmental lead is a persistent hazard that disproportionately affects children. Keys are known sources of lead exposure in children, but little is known about the specific lead content or frequency of lead in keys. We identified differences in lead concentration and frequency across items with different physical characteristics.

We compared the change in 24 circulating miRs from baseline to 2 years in a subset from DPP metformin intervention ( = 50) and placebo ( = 50) groups using Wilcoxon signed rank tests.

Data Sources: MEDLINE, Embase and Cochrane Library from 1 January 2020 to 19 May 2021.
Emerging evidence continues to demonstrate that disrupted insulin signaling and altered energy metabolism may play a key role underpinning pathology in neurodegenerative conditions. Intranasally administered insulin has already shown promise as a memory-enhancing therapy in patients with Alzheimer's and animal models of the disease. Intranasal drug delivery allows for direct targeting of insulin to the brain, bypassing the blood brain barrier and minimizing systemic adverse effects.

Metal chelators have been explored as a less well known approach to treatment. One chelator currently being developed is deferoxamine (DFO), administered via the intranasal (IN) route.
