A bread wheat line (N11) and a disomic 2D(2R) substitution triticale line were crossed and backrossed four times. At each step electrophoretic selection for the seeds that possessed, simultaneously, the complete set of high molecular weight glutenin subunits of N11 and the two high molecular weight secalins of rye, present in the 2D(2R) line, was carried out. Molecular cytogenetic analyses of the BCF generation revealed that the selection carried out produced a disomic addition line (2n = 44).

Background: Among wheat gluten proteins, the α-type gliadins are the major responsible for celiac disease, an autoimmune disorder that affects about 1% of the world population. In fact, these proteins contain several toxic and immunogenic epitopes that trigger the onset of the disease. The α-type gliadins are a multigene family, encoded by genes located at the complex Gli-2 loci.

Recently, electrophoretic analyses of waxy proteins in several hexaploid and tetraploid wheat accessions from worldwide collections have permitted the identification of new variants at the waxy loci, including allelic forms with different mobilities and partial null types. In this paper, the molecular characterization of mutated waxy loci in four bread wheat cultivars (two lacking the Wx-B1 and two lacking the Wx-D1 protein, respectively) and in four durum wheat cultivars (one lacking Wx-A1 and the remainder with Wx-B1 proteins showing different electrophoretic mobilities) was conducted by means of PCR, Southern and DNA sequence analyses. Three primer pairs were developed that identified six of the above-mentioned mutations and allowed their molecular description, providing a useful tool for further germplasm screening or marker assisted progeny selection in breeding programs involving the newly identified material.
