Background: The diagnosis of syndesmotic instability is challenging, and chronically unstable injuries can potentially lead to ankle arthritic degeneration. The objective of this cadaveric study was to utilize a 3-dimensional (3D) weight-bearing computed tomography (WBCT) distance mapping algorithm for the detection of subtle syndesmotic instability, induced by complete syndesmotic ligament sectioning and stressed by isolated axial load. We hypothesized that this algorithm would accurately detect subtle syndesmotic instability.

Article Synopsis
  • Transcatheter aortic valve implantation (TAVI) via transfemoral access is becoming more common in treating aortic valve disease, though access-related vascular complications still affect about 10% of procedures.
  • This study analyzed patients from Dutch and German hospitals who underwent TF-TAVI between 2017 and 2021, focusing on those who had occlusive complications (OC) versus hemorrhagic complications (HC) requiring further intervention.
  • Results showed that patients with OC had significantly smaller arterial diameters compared to those with HC, while those with HC had a higher tortuosity index, highlighting the need for careful preoperative imaging to tailor prevention strategies.

Embryogenesis entails dramatic shifts in mRNA translation and turnover that reprogram gene expression during cellular proliferation and differentiation. Codon identity modulates mRNA stability during early vertebrate embryogenesis, but how the composition of tRNA pools is matched to translational demand is unknown. By quantitative profiling of tRNA repertoires in zebrafish embryos during the maternal-to-zygotic transition, we show that zygotic tRNA repertoires are established after the onset of gastrulation, succeeding the major wave of zygotic mRNA transcription.

Article Synopsis
  • Deficiencies in the merging of paramesonephric ducts during embryonic development can lead to Müllerian malformations, which affect reproductive organs and can complicate cancer treatment.
  • A 41-year-old woman with atypical Pap smear results and HPV positivity was found to have cervical intraepithelial neoplasia and concurrent uterine and kidney anomalies, which complicated her diagnosis and treatment.
  • The case highlights the importance of personalized treatment plans for gynecological cancers in patients with anatomical anomalies, successfully showing that guideline-compliant therapy can be implemented for early-stage cervical cancer despite these challenges.

Article Synopsis
  • The Warburg effect in cancer cells leads to glucose being metabolized anaerobically, resulting in lactate buildup, which impacts chemotherapy response and DNA repair processes.
  • Lactate-induced lactylation of NBS1 is crucial for its role in DNA repair through homologous recombination and is influenced by specific enzymes that add or remove lactate.
  • High lactylation levels of NBS1 correlate with poor chemotherapy outcomes, and reducing lactate through specific inhibitors may enhance treatment effectiveness by improving DNA repair mechanisms.

Purpose: The receptor activator of nuclear factor kappa B (RANK) and its ligand (RANKL) have been shown to promote proliferation of the breast and breast carcinogenesis. The objective of this analysis was to investigate whether tumor-specific RANK and RANKL expression in patients with primary breast cancer is associated with high percentage mammographic density (PMD), which is a known breast cancer risk factor.

Methods: Immunohistochemical staining of RANK and RANKL was performed in tissue microarrays (TMAs) from primary breast cancer samples of the Bavarian Breast Cancer Cases and Controls (BBCC) study.

Optical projection tomography (OPT) is a three-dimensional mesoscopic imaging modality that can use absorption or fluorescence contrast, and is widely applied to fixed and live samples in the mm-cm scale. For fluorescence OPT, we present OPT implemented for accessibility and low cost, an open-source research-grade implementation of modular OPT hardware and software that has been designed to be widely accessible by using low-cost components, including light-emitting diode (LED) excitation and cooled complementary metal-oxide-semiconductor (CMOS) cameras. Both the hardware and software are modular and flexible in their implementation, enabling rapid switching between sample size scales and supporting compressive sensing to reconstruct images from undersampled sparse OPT data, e.

Humans lack the enzyme that produces the sialic acid N-glycolyl neuraminic acid (Neu5Gc), but several lines of evidence have shown that Neu5Gc can be taken up by mammalian food sources and replace the common human sialic acid N-acetyl neuraminic acid (Neu5Ac) in glycans. Cancer tissue has been shown to have increased the presence of Neu5Gc and Neu5Gc-containing glycolipids such as the ganglioside GM3, which have been proposed as tumor-specific antigens for antibody treatment. Here, we show that a previously described antibody against Neu5Gc-GM3 is binding to Neu5GC-containing gangliosides and is strongly staining different cancer tissues.

Trapped ion mobility spectrometry (TIMS) adds an additional separation dimension to mass spectrometry (MS) imaging, however, the lack of fragmentation spectra (MS) impedes confident compound annotation in spatial metabolomics. Here, we describe spatial ion mobility-scheduled exhaustive fragmentation (SIMSEF), a dataset-dependent acquisition strategy that augments TIMS-MS imaging datasets with MS spectra. The fragmentation experiments are systematically distributed across the sample and scheduled for multiple collision energies per precursor ion.
