The underlying physical chemistry behind electrochemically driven DNA denaturation is not well understood, and one possible mechanism is a change in local pH at the electrode surface. We demonstrate that by coimmobilization of p-mercaptobenozic acid at a dsDNA-functionalized electrode surface, it is possible to monitor both DNA denaturation and the local pH simultaneously using surface-enhanced Raman spectroscopy.

This structure is ideally suited to the investigation of the interfacial capacitance and properties of ionic liquids confined within small pores of the type found in the high surface area electrodes favoured for supercapacitors and batteries. Cyclic voltammetry experiments for BMIM-PF(6) show a large capacitance for the mesoporous Pt electrode, confirming that the ionic liquid fills the 2 nm pores.

During routine herd testing, Dairy Herd Improvement Association technicians collected milk samples from about 40 cows from each of 104 randomly selected Michigan dairy herds averaging ≥120 milking cows and 11,686 kg of milk/yr. Milk samples were analyzed for the presence of anti-BLV antibodies by ELISA, and herd- and lactation-specific estimates of BLV prevalence were computed to determine which were the most predictive of herd milk production, culling rate, and cow longevity (proportion of cows in their third or greater lactation).
View Article and Find Full Text PDFIn this paper, we report the novel finding that the cellular delivery efficiency of siRNAs or their mimic double-stranded (ds)DNA using layered double hydroxide (LDH) nanoparticles is dependent upon the nucleotide sequence. Efficacy of LDH-mediated delivery of four different siRNAs into cortical neurons and NIH 3T3 cells was found to vary widely (from 6 to 80%, and 2-11%, respectively). Our investigation into the formation of dsDNA-LDH complexes through monitoring the dsDNA:LDH mass ratio at the point of zero charge (PZC) indicated that the degree of intercalation of the individual dsDNA sequences into the LDH nanoparticles varied significantly.

Application of a voltage ramp can result in denaturation of dsDNA and strand separation. We show that the potential at which half of the surface immobilised duplexes denature (the melting potential, E(m)) directly correlates with the calculated nearest neighbour and experimental melting temperatures; T(m), for a duplex is solution. The results demonstrate that the electrochemical melting potential measures the stability of the dsDNA, and therefore existing nearest neighbour melting models can be utilized to design DNA probes with predictable electrochemical melting potentials for future assay applications.

For Ge(II) reagents the most promising results were obtained using [NBu(n)(4)][GeCl(3)]. Here, we show that Oncostatin M (Osm) and its receptor, OsmRβ, are both expressed in the subventricular zone (SVZ) and that in contradistinction to leukemia inhibitory factor and ciliary neutrophic factor, Osm directly inhibits the proliferation of adult NPCs as measured by a decreased level of neurosphere formation in vitro. Similarly, intraventricular infusion of Osm dramatically decreases the pool of NPCs in both the SVZ and the hippocampus.
