In this study, proteomic and metabolomic changes in leaves and roots of two barley (Hordeum vulgare L.) genotypes, with contrasting drought tolerance, subjected to water deficit were investigated. Our two-dimensional electrophoresis (2D-PAGE) combined with matrix-assisted laser desorption time of flight mass spectrometry (MALDI-TOF and MALDI-TOF/TOF) analyses revealed 121 drought-responsive proteins in leaves and 182 in roots of both genotypes.

Changes in physiological parameters (relative water content (RWC), biomass, water use efficiency (WUE), net photosynthetic yield (PN) and quantum yield of PSII (Fv/Fm)), in proline and sugar content, and expression profile of genes reported to be associated with the barley response to water deficit, including LEA genes, NHX1, Hsdr4, BLT101 and genes encoding transcription factors (HvDREB1, HvABF1, HvABI5 and HvZIP1), were analyzed in seedlings of nine barley genotypes subjected to a progressive increase in water deficit. Seedlings of all genotypes wilted when the soil water content (SWC) declined from 65% (control conditions) to 10% (severe drought conditions), but recovered turgor within a few hours of re-watering. However, when severe drought conditions were prolonged for a week, large differences in survival characteristics were observed between genotypes after re-watering.

These proteins have been proposed to be implicated in the active transmembrane transport of various secondary metabolites. Hairy roots and suspension cell cultures are commonly used in deciphering different problems related to the biochemistry and physiology of plant secondary metabolites. Here, we address about the issue of possible differences in the profiles of flavonoid compounds and their glycoconjugates derived from various plant materials grown in a standard culture media. We compared profiles of flavonoids isolated from seedling roots, hairy roots, and suspension root cell cultures of a model legume plant, Medicago truncatula.
