The plasma membrane has a complex organization that includes the polarized distribution of membrane proteins and lipids. Glycosylphosphatidylinositol-anchored proteins (GPI-APs) are ubiquitously expressed in eukaryotes and represent a functionally diverse, extensively remodeled, ER-derived group of proteins critical for the organization and function of the plasma membrane. Little is known about how the transport of incompletely remodeled GPI-APs to the plasma membrane affects cell function.
View Article and Find Full Text PDFThe pressure sensors on Mars rover Perseverance measure the pressure field in the Jezero crater on regular hourly basis starting in sol 15 after landing. The present study extends up to sol 460 encompassing the range of solar longitudes from ∼ 13°-241° (Martian Year (MY) 36). The data show the changing daily pressure cycle, the sol-to-sol seasonal evolution of the mean pressure field driven by the CO sublimation and deposition cycle at the poles, the characterization of up to six components of the atmospheric tides and their relationship to dust content in the atmosphere.

They are indicators of atmospheric turbulence and are an important lifting mechanism for the Martian dust cycle. Improving our understanding of dust lifting and atmospheric transport is key for accurate simulation of the dust cycle and for the prediction of dust storms, in addition to being important for future space exploration as grain impacts are implicated in the degradation of hardware on the surface of Mars.

We test this assumption by making the first comparison between Viking 1 and InSight surface pressure data, which were recorded 40 years apart. We describe a protocol to induce the synthesis of a GPI-anchored protein whereby EtNP is added to Man2 but not removed. By temporally manipulating the expression of Gpi7p, the enzyme that adds EtNP to Man2, in ted1Δ dcr2Δ cells prior to the expression of a canonical GPI-AP (mNeon-Gas1p), EtNP is attached to Man2 of de novo synthesized mNeon-Gas1p and cannot be removed.

Perseverance rover's novel environmental sensors and Jezero crater's dusty environment remedy this. GPI-AP remodeling events serve as quality assurance signatures, and complete remodeling of the anchor functions as a transport warrant. Glycosylphosphatidylinositol-anchored proteins (GPI-APs) undergo extensive posttranslational modifications and remodeling, including the addition and subsequent removal of phosphoethanolamine (EtNP) from mannose 1 (Man1) and mannose 2 (Man2) of the glycan moiety. Removal of EtNP from Man1 is catalyzed by Cdc1p, an event that has previously been considered to occur in the endoplasmic reticulum (ER). We establish that Cdc1p is in fact a /medial Golgi membrane protein that relies on the COPI coatomer for its retention in this organelle.

In addition to the geometric variation in illumination on seasonal and diurnal timescales, the data are influenced by dust suspended in the atmosphere and deposited on the solar panels. Although no dust devils have been detected by InSight's cameras, brief excursions in solar array currents suggest that at least some of the vortices detected by transient pressure drops are accompanied by dust.

However, in sum the current compendium of mechanisms cannot account for the localization of all Golgi membrane proteins, and this is particularly the case for proteins such as the glycosyltransferases. Here we report on the geology and subsurface structure of the landing site to aid in situ geophysical investigations. InSight landed in a degraded impact crater in Elysium Planitia on a smooth sandy, granule- and pebble-rich surface with few rocks.

To understand the processes that control the interaction between surface and atmosphere on Mars, not only the mean winds, but also the turbulent boundary layer, the fluxes of momentum, heat and molecular constituents between surface and atmosphere must be measured. Terrestrially this is done with acoustic anemometers, but the low density atmosphere on Mars makes it challenging to adapt such an instrument for use on Mars.

We determined dissolved methane concentrations (n = 36) and δC and δH values (n = 31) in methane and groundwater from the 46,000-km Gippsland Basin in southeast Australia. The basin contains important water supply aquifers and is a potential target for future unconventional gas development.

The composition and length of a SNARE's transmembrane domain (TMD) is also an indicator for their steady-state distribution in cells. The evolutionary conservation of the SNARE TMD, together with the strict requirement of this feature for membrane fusion in biochemical studies, implies that the TMD represents an essential protein module.
View Article and Find Full Text PDFWe recently identified a new COPI-interacting KXD/E motif in the C-terminal cytosolic tail (CT) of Arabidopsis endomembrane protein 12 (AtEMP12) as being a crucial Golgi retention mechanism for AtEMP12. This KXD/E motif is conserved in CTs of all EMPs found in plants, yeast, and humans and is also present in hundreds of other membrane proteins. Here, by cloning selective EMP isoforms from plants, yeast, and mammals, we study the localizations of EMPs in different expression systems, since there are contradictory reports on the localizations of EMPs.
