Article Synopsis
  • The study investigates the effects of the HIV protease inhibitor nelfinavir, boosted with ritonavir, on excessive cell death (apoptosis) in mouse models of nonviral diseases.
  • Results show that protease inhibitors significantly reduce apoptosis and improve tissue health, function, and recovery in various conditions such as hepatitis, shock, and stroke.
  • The study concludes that these inhibitors block apoptosis by protecting mitochondrial integrity and preventing the function of a specific mitochondrial pore complex.

It has been proposed that antiretroviral therapies (ART) possess both antiviral and immunomodulatory activities when used in HIV infected patients. Few studies have addressed whether these putative immunomodulatory effects are also seen in HIV negative patients, for example, when used for post exposure prophylaxis (PEP). We chose to evaluate immunologic function in HIV negative patients who received Nelfinavir and Combivir (AZT and 3TC) as PEP.

Infection of T cells with HIV-1 induces apoptosis and modulates apoptosis regulatory molecules. Similar effects occur following treatment of cells with individual HIV-1 encoded proteins. While HIV-1 protease is known to be cytotoxic, little is known of its effect on apoptosis and apoptosis regulatory molecules.
