A new technique for the assay of carbohydrates is described in which separation and quantification of neutral saccharides, aminosaccharides, glycuronic acids, and disaccharides may be accomplished in less than 50 min of total run time. This method involves optimized anion-exchange liquid chromatography coupled with integrated pulse amperometric detection. Complex carbohydrates from various sources, including dietary supplements, were hydrolyzed in a dilute solution of trifluoroacetic acid, freeze-dried, and reconstituted in water containing 2-deoxygalactose as the internal standard.

View Article and Find Full Text PDFAloe polymannose (AP), a high mannose biological response modifier (BRM) purified from the Aloe barbadensis Miller plant, was tested for activity in enhancing antibody titres against coxsackievirus B3 (CVB3) and CVB3-induced myocarditis in murine models of the disease. Inoculation of mice with AP over a range of three nontoxic doses and in varying schedules did not reduce virus titres in heart tissues or ameliorate virus-induced cardiopathological alterations during acute disease. However, this BRM was found to significantly enhance titres of anti-CVB3 antibodies produced during acute infection of three strains of mice with CVB3.
View Article and Find Full Text PDFA complex glyconutritional (GN) mixture of mono-, di-and polysaccharides was investigated to assess its capacity to protect two different types of rodent cells, rat hepatocytes and mouse splenocytes, from depletion of glutathione by a sulfhydryl-reactive mycotoxin, patulin, or by coxsackievirus B3 (CVB3) infection, respectively. Rat hepatocytes were treated with the GN mixture in vitro or received carrier medium only prior to treatment with patulin. When treated with the GN mixture prior to patulin exposure hepatocytes demonstrated protection against depletion of intracellular reduced glutathione (GSH).

These animals had recurring disease that had failed previous treatment, a poor prognosis for survival, or both. Following four to seven weekly acemannan treatments, tumor shrinkage occurred in four (greater than 50%; n = 2) of 12 animals, with tumors accessible to measurement.
