Background: The burden of pneumococcal diseases remains high in Japan. Pn-MAPS24v is a novel MAPS-based vaccine containing complexes of 24 serotype-specific polysaccharides (PS), non-covalently coupled with fusion protein 1 (CP1). This study evaluated the safety and immunogenicity of different dose levels of Pn-MAPS24v, administered in Japanese adults either subcutaneously (SC) or intramuscularly (IM).

Article Synopsis
  • The phase II STARLIGHT study aimed to evaluate the effectiveness and safety of fezolinetant for treating vasomotor symptoms (VMS) in Japanese women undergoing menopause and to determine the best dosage for future studies.
  • The study involved 147 perimenopausal and postmenopausal women, who were randomly assigned to receive either 15 mg or 30 mg of fezolinetant or a placebo daily for 12 weeks, with changes in VMS frequency tracked through daily diaries.
  • Results showed that both doses of fezolinetant significantly reduced VMS frequency compared to placebo by week 8, with the treatment being well tolerated and showing no major safety concerns.

Article Synopsis
  • Basket trials, which include multiple cancer types, allow researchers to utilize information from different groups (or "baskets") to improve trial efficiency, but treatment effects can vary significantly across these types.
  • Current basket trial designs often assume that cancer types can be treated as exchangeable, which may not always be valid; this study presents a new model to account for treatment effect differences among cancer types.
  • The simplified Constrained Hierarchical Bayesian Model for Latent Subgroups (CHBM-LS) enhances information sharing within subgroups and has shown improved performance in real trials and simulations, offering greater statistical power and better management of type I errors compared to existing methods.

Unlabelled: We assessed the efficacy and safety of fidaxomicin, a narrow-spectrum macrocyclic antibiotic, for treating inpatients with Clostridioides (Clostridium) difficile infection (CDI) in Japan. The objective was to demonstrate the non-inferior efficacy of fidaxomicin versus vancomycin. This Phase III, vancomycin-controlled, double-blind, parallel-group study enrolled adults with CDI.

Cu nanoparticles (NPs) are widely used in numerous applications because of their unique optical, catalytic, and electrical properties. However, the synthesis of monodisperse Cu NPs often involves harmful reducing agents under an inert atmosphere in a solution with low Cu ion concentrations. In this study, monodisperse Cu NPs were synthesized by the thermal decomposition of an oleylamine-coordinated Cu oxalate (Cu(ox)) complex (OA-Cu(ox)) in oleylamine in air with no reducing agent.
