Klotho deficiency is prevalent in various chronic kidney diseases. Although klotho is known to bind transforming growth factor β (TGFβ) receptor 1 to antagonize renal fibrosis, TGFβ also maintains regulatory T cells with inducing forkhead box protein P3 (FOXP3). Despite significant advances in the treatment of cutaneous melanoma (hereafter melanoma), the prognosis remains less favorable due to therapeutic resistance, which is presumably linked to epigenetic dysregulation. We hypothesized that the histone lysine demethylase KDM4B could play a pivotal role in controlling therapy-resistant melanoma. To validate our hypothesis, we retrieved RNA sequencing data from the Gene Expression Omnibus (GEO) and The Cancer Genome Atlas (TCGA) program and observed upregulation of KDM4B in both primary and metastatic melanoma, which was associated with poor survival.

CD146/MCAM has garnered significant attention for its potential contribution to cardiovascular disease; however, the transcriptional regulation and functions remain unclear. To explore these processes regarding cardiomyopathy, we employed doxorubicin, a widely used stressor for cardiomyocytes. Our in vitro study on H9c2 cardiomyoblasts highlights that, besides impairing the fatty acid uptake in the cells, doxorubicin suppressed the expression of fatty acid binding protein 4 (Fabp4) along with the histone deacetylase 9 (Hdac9), bromodomain and extra-terminal domain proteins (BETs: Brd2 and Brd4), while augmented the production of CD146/MCAM.

A 58-year-old woman with a history of morbid obesity, asthma, and prior warfarin use presented to the hospital with shortness of breath and a three-month history of painful, ulcerated ulcers with retiform purpura of her bilateral distal extremities. A punch biopsy specimen demonstrated focal necrosis and hyalinization of the adipose tissue with subtle arteriolar calcium deposition, findings consistent with calciphylaxis. We discuss the presentation of non-uremic calciphylaxis and review the risk factors, pathophysiology, and interdisciplinary management approach of this rare disease.

We previously reported that silencing of the gene, which encodes the (pro)renin receptor [(P)RR], significantly reduced Wnt/β-catenin-dependent development of pancreatic ductal adenocarcinoma (PDAC). Here, we examined the effects of a panel of blocking mAbs directed against the (P)RR extracellular domain on proliferation of the human PDAC cell lines PK-1 and PANC-1 and We observed that four rat anti-(P)RR mAbs induced accumulation of cells in the G-G-phase of the cell cycle and significantly reduced proliferation concomitant with an attenuation of Wnt/β-catenin signaling. Systemic administration of the anti-(P)RR mAbs to nude mice bearing subcutaneous PK-1 xenografts significantly decreased tumor expression of active β-catenin and the proliferation marker Ki-67, and reduced tumor growth.

The topological characteristics of waves in elastic structures are determined by the geometric phase of waves and, more specifically, by the Berry phase, as a characterization of the global vibrational behavior of the system. A computational procedure for the numerical determination of the geometrical phase characteristics of a general elastic structure is introduced: the spectral analysis of amplitudes and phases method. Molecular dynamics simulation is employed to computationally generate the band structure, traveling modes' amplitudes and phases, and subsequently the Berry phase associated with each band of periodic superlattices.

Solitary waves (SWs) are generated in monoatomic (homogeneous) lightly contacting spherical granules by an applied input force of any time-variation and intensity. We consider finite duration shock loads on one-dimensional arrays of granules and focus on the transition regime that leads to the formation of SWs. Based on geometrical and material properties of the granules and the properties of the input shock, we provide explicit analytic expressions to calculate the peak value of the compressive contact force at each contact point in the transition regime that precedes the formation of a primary solitary wave.
