Background: Diagnosis of allergies is mostly based on the patient's clinical history and allergen provocation tests. Determination of specific IgE (sIgE) profiles can be performed to support allergy diagnosis. This is commonly done in vivo by the skin prick test or in vitro with automated systems.

Background: The clinical significance of newly available platforms for specific IgE measurement must be evaluated. However, data are lacking for NOVEOS (Hycor), especially for food allergens.

Objective: We compared the technical and clinical performance of two platforms (ImmunoCAP and NOVEOS) to measure specific IgE to 10 food allergens.

Article Synopsis
  • Systemic mastocytosis (SM) is associated with vertebral osteoporosis (OP) and fractures, but the relationship between OP and mast cell abnormalities is unclear.
  • A study compared OP characteristics in patients with cutaneous mastocytosis (CM) and monoclonal mast cell activation syndrome (MMAS) to those with nonadvanced SM, finding that CM patients had lower rates of OP and vertebral fractures.
  • The results suggest that OP in CM patients differs in management and implications from that in MMAS and nonadvanced SM, indicating the need for further research to improve understanding and treatment.

Background: Type I interferon (IFN-I) production by plasmacytoid dendritic cells (pDCs) occurs during viral infection, in response to Toll-like receptor 7 (TLR7) stimulation and is more vigorous in females than in males. Whether this sex bias persists in ageing people is currently unknown. In this study, we investigated the effect of sex and aging on IFN-α production induced by PRR agonist ligands.

Accreditation of an in vitro diagnostic assay according to the NF/EN/ISO 15189 standard requires to analyze its technical performance before implementation for routine use, and annually when reviewing effectiveness of quality controls. Performance is evaluated through repeatability, intermediate fidelity, accuracy and uncertainty of measurement. The coefficients of variation (CV) of the intra-assay and inter-assay precision tests must be compared with those of "peers" (results from laboratories employing the same method) and also with those obtained with "all methods", i.

Background: Mastocytosis is characterized by the accumulation/proliferation of abnormal mast cells. The frequency of isolated cutaneous involvement in adults with mastocytosis has not been fully determined. The main objective of our study was to assess the frequency of isolated cutaneous mastocytosis (CM) in adults with mastocytosis skin lesions.

Unlabelled: Little is known about osteoporosis in mast cell disorders (MCDs) not related to systemic mastocytosis. We described osteoporosis and fractures in MCDs and showed that systemic mastocytosis was the only studied MCDs associated with osteoporotic vertebral fractures.

Introduction: To describe osteoporosis (OP) and fragility fractures in mast cell disorders (MCDs).

Background: Peach is a common elicitor of food allergic reactions. Peach-induced immediate reactions may occur as benign pollen-food syndromes, usually due to birch pollen-related PR-10 cross-reactivity in temperate climates, and as potentially severe primary food allergies, predominantly related to nsLTP Pru p 3 in Mediterranean regions. The newly described peach allergen Pru p 7 has gained recent attention as a potential peach allergy severity marker.

There is no therapeutic agent approved in cutaneous mastocytosis and mast cell activation syndrome. We report the efficacy of hydroxychloroquine in four patients with cutaneous mastocytosis (n = 2) and mast cell activation syndrome (n = 2). We show that this molecule reduces the long-term survival of primary human mast cells, interferes with lysosome function and leads to the accumulation of non-functional tryptase in the mast cell granules.

Article Synopsis
  • A study analyzed blood cell types in 256 healthy individuals aged 19 to 96, examining how age, sex, and cytomegalovirus (CMV) impacted monocyte and dendritic cell populations using flow cytometry. !* -
  • Findings showed that classical, intermediate, and nonclassical monocyte levels increased slightly with age, while myeloid and plasmacytoid dendritic cell counts remained stable; sex had minimal impact on these values. !* -
  • Notably, CMV status influenced the decrease in plasmacytoid dendritic cells with age, suggesting that lifelong CMV infection may affect immune cell circulation, though it didn't significantly change the overall numbers of myeloid dendritic

Article Synopsis
  • Systemic mastocytosis (SM) is a condition where mast cells accumulate in various tissues, often leading to weak bones and fragility fractures (FF), which was the main focus of the study.
  • The study analyzed data from 89 SM patients to evaluate risk factors for FF, including age, bone density, symptoms, and bone marrow tryptase (BMT) levels.
  • Results showed that older age, low femoral T-score, and high BMT levels are significant indicators for predicting FF in SM patients, suggesting BMT could be a useful biomarker for these fractures.

The data presented in this paper are reference ranges for frequencies of thirty-eight subpopulations of T, B and NK lymphocytes, established from a cohort of 253 healthy blood donors aged from 19 to 67. When relevant, the influence of age or sex was taken into account to calculate these reference values. This article is related to the research article entitled "Influence of age, sex and HCMV-serostatus on blood lymphocyte subpopulations in healthy adults" (Apoil et al.

Article Synopsis
  • - We analyzed 38 different subpopulations of T, B, and NK lymphocytes in 253 healthy blood donors to see how factors like age, sex, and HCMV infection affect these cells.
  • - Aging was found to increase specific lymphocyte types, such as CD8+ EMRA T cells and Tregs, while older individuals also showed a rise in HLA-DR+ Tregs and CD4+ T lymphocytes.
  • - Sex differences were noted, with females having higher counts of CD4+ T cells, and HCMV-positive individuals had more CD8+ EMRA memory T cells and a unique pattern of terminally differentiated CD4+ T cells.

Overlap chronic graft-versus-host disease (GVHD) associates both features of acute and chronic GVHD. Trigger factors for chronic GVHD are unclear. We describe two patients who received allogenic haematopoietic stem-cell transplantation, and who later developed overlap chronic GVHD after sun exposure.

Background: Telangiectasia macularis eruptiva perstans (TMEP) has not been fully characterized.

Objective: We sought to estimate the frequency and clinical characteristics of TMEP in a cohort of adult patients with cutaneous mastocytosis, and to assess the presence of systemic involvement.

Methods: We included all consecutive patients evaluated for cutaneous mastocytosis in 2 centers: the Mastocytosis Competence Center of the Midi-Pyrénées from May 2006 to December 2013, and the French Reference Center for Mastocytosis from January 2008 to September 2013.

Background: Mastocytosis is difficult to diagnose, especially when systemic mast cell activation symptoms are not present or involve only one extracutaneous organ.

Objective: The main objective was to evaluate the accuracy of the bone marrow tryptase level in the diagnosis of systemic mastocytosis in patients with a clinical suspicion of mastocytosis.

Methods: We included all adult patients evaluated in our centre between December 2009 and 2013 for suspected mastocytosis as part of a standardized procedure and who had a bone marrow and serum tryptase assay on the same day.
