Article Synopsis
  • Japanese patients with very high-risk cutaneous squamous cell carcinomas (cSCCs) experience higher rates of relapse and disease-specific death compared to those with just high-risk cSCCs, highlighting the need for effective prognosis prediction.
  • The study analyzed data from 424 Japanese patients to assess the effectiveness of a new prognostic tool called the Japanese Risk Factor Scoring System (JARF scoring) against other established systems like the Brigham and Women's Hospital staging and the NCCN risk factors.
  • Results indicated that the JARF scoring system is particularly effective in predicting poor outcomes such as local recurrence, lymph node metastasis, disease-specific death, and overall survival in patients with very high-risk cSCC in

Mild acidification caused by transcutaneous administration of carbon dioxide (CO ) has been reported to improve some epidermal skin impairments, such as desquamation and inflammation; however, its effects on dermal tissue remain unclear. Here, we examined the effect and mechanism of mild acidity on extracellular matrix (ECM) protein production in normal human dermal fibroblasts (NHDFs). To achieve this, the skin permeability of CO and its effect on intradermal pH were evaluated by treating reconstructed human skin equivalents (HSEs) with a CO -containing formulation.

Chronic myeloid leukemia (CML) is a well-characterized oncological disease in which virtually all patients possess a translocation (9;22) that generates the tyrosine kinase protein. This translocation represents one of the milestones in molecular oncology in terms of both diagnostic and prognostic evaluations. The molecular detection of the transcription is a required factor for CML diagnosis, and its molecular quantification is essential for assessing treatment options and clinical approaches.

Background/aim: A recent clinical trial indicated the usefulness of local radiation therapy of the prostate in patients with low-volume metastatic prostate cancer. High-dose-rate brachytherapy (HDR-BT) is used mainly for high-risk, localized, and locally advanced cases. However, few studies exist on the efficacy of HDR-BT and external beam radiation therapy (EBRT) for metastatic prostate cancer.

Background/aim: We retrospectively investigated the effect of a biologically effective dose (BED) of Low-dose rate brachytherapy (LDR-BT) and its possible interaction with androgen deprivation therapy (ADT) during LDR-BT treatment for intermediate-risk prostate cancer (PCa).

Patients And Methods: A total of 693 patients with localized, intermediate-risk PCa, who underwent LDR-BT with or without supplemental external beam radiotherapy, were included in this study. We stratified patients into two groups according to BED (<180 Gy2, lower BED group; ≥180 Gy2, higher BED group) and evaluated the effect of ADT duration on the oncological outcomes of each group.

Background/aim: Dendritic cells (DCs) are difficult to evaluate in lung regional lymph nodes because of region-specific structures, such as abundant trabeculae connecting the medullary and subcapsular sinuses, the latter of which contains few anthracotic macrophages. Therefore, DC-specific intercellular adhesion molecule-3-grabbing non-integrin (DCsign)-positive DCs and CD68-positive macrophages are unlikely to show a typical distribution. The present study therefore explored quantitative factors connecting the nodal DC morphology to the patient outcome.

Amyotrophic lateral sclerosis (ALS) is a neurodegenerative disorder characterized by the degeneration of motor neurons. Although repeat expansion in C9orf72 is its most common cause, the pathogenesis of ALS isn't fully clear. In this study, we show that repeat expansion in LRP12, a causative variant of oculopharyngodistal myopathy type 1 (OPDM1), is a cause of ALS.

Development of a universal influenza vaccine that can provide robust and long-lasting protection against heterologous infections is a global public health priority. A variety of vaccine antigens are designed to increase the antigenicity of conserved epitopes to elicit cross-protective antibodies that often lack virus-neutralizing activity. Given the contribution of antibody effector functions to cross-protection, adjuvants need to be added to modulate antibody effector functions as well as to enhance antibody quantity.

The incidence of seasonal infections due to respiratory viruses other than severe acute respiratory coronavirus virus 2 (SARS-CoV-2) has declined due to heightened public infection prevention measures against coronavirus disease 2019 (COVID-19). We describe an outbreak of human coronavirus OC43 infection that occurred at a long-term care facility and whose clinical features were indistinguishable from COVID-19.

Background And Objective: The prognosis of advanced urothelial carcinoma (aUC) is poor. To date, the gold standard of treatment for patients with aUC has been cisplatin-based chemotherapy. More recently, immune checkpoint inhibitors (ICIs) have been widely used for such patients, which has led to an improvement in their prognosis.

Cell-to-cell spreading of misfolded α-synuclein (αSYN) is supposed to play a key role in the pathological progression of Parkinson's disease (PD) and other synucleinopathies. Receptor-mediated endocytosis has been shown to contributes to the uptake of αSYN in both neuronal and glial cells. To determine the receptor involved in αSYN endocytosis on the cell surface, we performed unbiased, and comprehensive screening using a membrane protein library of the mouse whole brain combined with affinity chromatography and mass spectrometry.

Article Synopsis
  • iPSC-based drug discovery led to a clinical trial investigating ropinirole for treating sporadic ALS, involving 20 participants over 24 weeks to assess safety and effects.
  • The results showed that while muscle strength and daily activities were stable, the primary functional measure (ALSFRS-R) did not significantly improve compared to the placebo group during the double-blind period.
  • In an open-label extension, the ropinirole group demonstrated a significant slowdown in ALSFRS-R decline and extended disease-progression-free survival, but the study faced challenges like small sample size and participant dropouts, necessitating further research.

Although the risks faced by passengers in near-side lateral collisions are understood, and despite the presence of side airbags for injury prevention, passengers involved in far-side lateral collisions also suffer serious and fatal injuries. The objective of this study was to determine the independent predictive factors of fatality of motor vehicle passengers involved in far-side lateral collisions. Using 2010 records from the National Automotive Sampling System/Crashworthiness Data System (NASS/CDS), we selected 86 fatal and 325 non-fatal passengers with an Abbreviated Injury Scale (AIS) score of 2 or more.

Oxyhydrides are promising compounds as supports for ammonia synthesis catalysts because they suppress hydrogen poisoning on the catalyst surface and enhance the ammonia synthesis activity. Herein, we developed a facile method for preparing BaTiOH, a perovskite oxyhydride, on a TiH surface the conventional wet impregnation method using TiH and Ba hydroxide. Scanning electron microscopy and high-angle annular dark-field scanning transmission electron microscopy observations revealed that BaTiOH crystallized as nanoparticles of 100-200 nm on the TiH surface.

Article Synopsis
  • Abdominal compartment syndrome (ACS) is a serious complication that can occur after blunt abdominal trauma, requiring quick diagnosis and surgery for the best outcomes.
  • A study using data from the Japan Trauma Data Bank found that among 294,274 patients, 150 (1.3%) developed ACS, with those affected showing more severe injuries, including higher rates of abdominal organ damage, vascular and pancreatic injuries, and increased mortality rates (51.1% vs. 26.0% in controls).
  • The analysis identified two key risk factors for ACS: a greater number of injured abdominal organs and specific pancreatic injuries, both of which independently increase the likelihood of developing the syndrome.
