Article Synopsis
  • Fumarate hydratase (FH)-deficient renal cell carcinomas are uncommon but aggressive tumors linked to mutations in the FH gene, displaying significant morphological variability and often spreading rapidly to distant organs.
  • In a study of 11 primary tumors and 7 metastases, most metastases were found in abdominal lymph nodes and the peritoneum, with tissue examination revealing diverse structures that can differ from the original tumors.
  • The research highlighted that while 60% of primary tumors showed PD-L1 expression, there were no identified EGFR mutations, suggesting the potential for targeted immunotherapy and the importance of accurate diagnosis to differentiate these tumors from other conditions.

Eyelid melanoma (EM) is a malignant neoplasm accounting for around 1% of eyelid malignancies. Because of its rarity, most of our knowledge of EM is currently based on studies of cutaneous melanomas located elsewhere. Accordingly, this study aimed to specifically evaluate EM characteristics, management strategies, and prognosis.

  • Undifferentiated sarcomatoid carcinoma (USC) of the pancreas is a rare and aggressive form of pancreatic cancer, characterized by at least 80% sarcomatoid cells and poor patient prognosis.
  • A study of 10 patients revealed similar genetic alterations to conventional pancreatic ductal adenocarcinoma (PDAC), notably frequent mutations in KRAS and TP53, but with less alteration in SMAD4 and some cases showing unique KRAS amplification.
  • The findings highlight the aggressive nature of pancreatic USC, its distinct molecular profile, and the presence of potential treatment targets, despite having limited druggable alterations compared to conventional PDAC.

Ki-67 assessment is a key step in the diagnosis of neuroendocrine neoplasms (NENs) from all anatomic locations. Several challenges exist related to quantifying the Ki-67 proliferation index due to lack of method standardization and inter-reader variability. The application of digital pathology coupled with machine learning has been shown to be highly accurate and reproducible for the evaluation of Ki-67 in NENs.
