Article Synopsis
  • T cells are crucial in the development of systemic lupus erythematosus (SLE), and chronic signaling can lead to T cell exhaustion, reducing their tissue damage capacity.
  • Researchers tested an anti-TCRβ monoclonal antibody (mAb) in SLE mouse models, finding that it significantly increased survival rates and reduced kidney damage compared to controls.
  • The treatment also altered T cell profiles, increasing regulatory T cells and inducing an exhaustion-like state in CD4 T cells, suggesting that promoting T cell exhaustion could be a beneficial therapeutic strategy for SLE.

Neuroblastoma (NB) is a pediatric tumor of the peripheral nervous system. Approximately 80% of relapsed NB show RAS-MAPK pathway mutations that activate ERK, resulting in the promotion of cell proliferation and drug resistance. Ulixertinib, a first-in-class ERK-specific inhibitor, has shown promising antitumor activity in phase 1 clinical trials for advanced solid tumors.

T cell stemness and exhaustion coexist as two key contrasting phenomena during chronic antigen stimulation, such as infection, transplant, cancer, and autoimmunity. T cell exhaustion refers to the progressive loss of effector function caused by chronic antigen exposure. Exhausted T (T) cells highly express multiple inhibitory receptors and exhibit severe defects in cell proliferation and cytokine production.
