Article Synopsis
  • FLNA is a protein crucial for cytoskeletal structure and signaling, with its C-terminal region playing a vital role in homodimer formation and interactions with other proteins.
  • A study examined mutations in FLNA, particularly in male patients, revealing varying phenotypes, including typical female presentations of periventricular heterotopia (PH) and a unique mild presentation in brothers caused by a specific mutation.
  • Findings indicate that FLNA mutations can disrupt dimerization and lead to diverse phenotypic outcomes, highlighting the complexity of genotype-phenotype relationships for conditions associated with FLNA.

The squash (Cucurbita maxima) phloem exudate-expressed aspartic proteinase inhibitor (SQAPI) is a novel aspartic acid proteinase inhibitor, constituting a fifth family of aspartic proteinase inhibitors. However, a comparison of the SQAPI sequence to the phytocystatin (a cysteine proteinase inhibitor) family sequences showed approximately 30% identity. Modeling SQAPI onto the structure of oryzacystatin gave an excellent fit; regions identified as proteinase binding loops in cystatin coincided with regions of SQAPI identified as hypervariable, and tryptophan fluorescence changes were also consistent with a cystatin structure.
