The progressive decline of dopaminergic neurons in Parkinson's disease (PD) has been linked to an imbalance in energy and the failure of mitochondrial function. AMP-activated protein kinase (AMPK), the major intracellular energy sensor, regulates energy balance, and damage to nigral dopaminergic neurons induced by 6-hydroxydopamine (6-OHDA) is exacerbated in the absence of AMPK activity. This study aimed to examine the potential therapeutic advantages of AdipoRon, an AMPK activator, on motor function and mitochondrial homeostasis in a 6-OHDA-induced PD model.

Article Synopsis
  • This study investigates the effects of the adiponectin receptor agonist, AdipoRon (Adipo), on motor function and cognitive performance in a rat model of Parkinson’s disease (PD) induced by 6-hydroxydopamine (6-OHDA).
  • Chronic treatment with Adipo improved both motor impairments and memory functions in hemiparkinsonian rats, particularly at doses of 1 and 10 μg, while also being compared to levodopa treatment.
  • Biochemical analysis revealed that Adipo reduced oxidative stress and increased levels of brain-derived neurotrophic factor (BDNF) and synaptic proteins, suggesting a mechanism for its neuroprotective effects.

Article Synopsis
  • Psoriasis is driven by immune cell interactions, with a focus on S1PR2, which hasn’t been thoroughly explored before, despite other S1PR antagonists showing promise in treatment.
  • Using techniques like spatial transcriptomics and flow cytometry, researchers studied how S1PR2 impacts immune responses in a mouse model of psoriasis.
  • The findings indicate that S1PR2 in keratinocytes helps regulate inflammation by inhibiting Th17 cell recruitment, suggesting that activating S1PR2 might offer a new treatment strategy for psoriasis.

Depression and anxiety are prevalent neuropsychiatric conditions among patients with Parkinson's disease (PD), which may manifest prior to motor symptoms. As levodopa, a prominent treatment for PD motor symptoms, provides few benefits for mood-related abnormalities, tackling non-motor symptoms is particularly important. AdipoRon (Ad), an adiponectin agonist, has demonstrated neuroprotective effects by suppressing neuroinflammatory responses and activating the AMPK/Sirt-1 signaling pathway.

Mast cells (MCs) are tissue-resident immune cells of a hematopoietic origin that play vital roles in innate and adaptive immunity. Human MCs can be isolated and differentiated from various tissue sources, including cord blood, when supplemented with cytokines such as stem cell factor, interleukin 3, and interleukin 6. Our current research study has shown significant differences in the marker expressions of human cord blood-derived mast cells (hCBMCs) based on donor dependency and the type of medium used for culturing and differentiation.

Introduction: Extracts and compounds isolated from hemp (Cannabis sativa) are increasingly gaining popularity in the treatment of a number of diseases, with topical formulations for dermatological conditions leading the way. Phytocannabinoids such as ( )-cannabidiol, ( )-cannabinol and ( )-Δ9-tetrahydrocannabivarin (CBD, CBN, and THCV, respectively), are present in variable amounts in the plant, and have been shown to have mostly anti-inflammatory effects both in vitro and in vivo, albeit dominantly in murine models. The role of phytocannabinoids in regulating responses of dendritic cells (DCs) remains unclear.

Activation and degranulation of mast cells (MCs) is an essential aspect of innate and adaptive immunity. Skin MCs, the most exposed to the external environment, are at risk of quickly degranulating with potentially severe consequences. Here, we define how MCs assume a tolerant phenotype via crosstalk with dermal fibroblasts (dFBs) and how this phenotype reduces unnecessary inflammation when in contact with beneficial commensal bacteria.

Langerhans cells (LCs) are the sole professional antigen-presenting cell normally found in the human epidermal compartment. Research into their physiological role is hindered by the fact that they are invariably activated during isolation from the skin. To overcome this challenge, we turned to a monocyte-derived LC (moLC) model, which we characterized with RNA sequencing, and compared the transcriptome of moLCs with that of donor-matched immature dendritic cells.

Surfactant-enhanced aquifer remediation (SEAR) is an appropriate method for DNAPL-contaminated aquifer remediation; However, due to the high cost of the SEAR method, finding the optimal remediation scenario is usually essential. Embedding numerical simulation models of DNAPL remediation within the optimization routines are computationally expensive, and in this situation, using surrogate models instead of numerical models is a proper alternative. Ensemble methods are also utilized to enhance the accuracy of surrogate models, and in this study, the Stacking ensemble method was applied and compared with conventional methods.

Photodamage-induced and viral keratitis could benefit from treatment with novel nonsteroid anti-inflammatory agents. Therefore, we determined whether human corneal epithelial cells (HCECs) express members of the endocannabinoid system (ECS), and examined how the endocannabinoid anandamide (AEA, N-arachidonoyl ethanolamine) influences the Toll-like receptor 3 (TLR3) agonism- or UVB irradiation-induced inflammatory response of these cells. Other than confirming the presence of cannabinoid receptors, we show that endocannabinoid synthesizing and catabolizing enzymes are also expressed in HCECs in vitro, as well as in the epithelial layer of the human cornea in situ, proving that they are one possible source of endocannabinoids.
