L-arginine: glycine amidinotransferase (AGAT) gained academic interest as the rate-limiting enzyme in creatine biosynthesis and its role in the regulation of creatine homeostasis. Of clinical relevance is the diagnosis of patients with AGAT deficiency but also the potential role of AGAT as therapeutic target for the treatment of another creatine deficiency syndrome, guanidinoacetate N-methyltransferase (GAMT) deficiency. Applying a stable isotope-labeled substrate method, we utilized ARG 15N (ARG-δ2) and GLY 13C15N (GLY-δ3) to determine the rate of 1,2-13C,15N guanidinoacetate (GAA-δ5) formation to assess AGAT activity in various mouse tissue samples and human-derived cells.

The LuX-Valve Plus is a novel radial force-independent orthotopic transjugular transcatheter tricuspid valve replacement device proven to be effective in TR reduction. We describe preprocedural assessment for eligibility and procedural planning by means of computed tomography as well as procedural steps of device implantation under multimodality imaging guidance including transesophageal echocardiography (TEE), intracardiac echocardiography (ICE), and fluoroscopy: steering into the right ventricle, leaflet capture by graspers, fine valve adjustment, and septal anchor deployment. Potential pitfalls are avoided by achieving optimal alignment by means of TEE multiplanar reconstruction to steer the delivery system, and using mid-esophageal and deep gastric views of 3-dimensional TEE and supplementary ICE to visualize the graspers for leaflet capture, especially when TEE imaging is technically challenging.

Oxygen vacancies and their correlation with the nanomagnetism and electronic structure are crucial for applications in dilute magnetic semiconductors design applications. Here, we report on cobalt single atom-incorporated titanium dioxide (TiO) monodispersed nanoparticles synthesized using a thermodynamic redistribution strategy. Using advanced synchrotron-based X-ray techniques and simulations, we find trivalent titanium is absent, indicating trivalent cations do not influence ferromagnetic (FM) stability.

Background: Targeting the enzyme L-Arginine:glycine amidinotransferase (AGAT) to reduce the formation of guanidinoacetate (GAA) in patients with guanidinoacetate methyltransferase (GAMT) deficiency, we attempted to identify drugs for repurposing that reduce the expression of AGAT via transcriptional inhibition.

In this article, we describe a novel case of SARS-CoV-2-associated-hemophagocytic lymphohistiocytosis (HLH) complicated by posterior reversible encephalopathy syndrome (PRES). Initially diagnosed with multisystem inflammatory response in children (MIS-C), the patient received a large corticosteroid dose days before the onset of neurological symptoms. After developing PRES, the patient was treated with antihypertensives, antiepileptics, dexamethasone, and anakinra, leading to neurologic normalization.
