Effect of synthetic analogues of atrial natriuretic factor (ANF) on proliferative response and natural cytotoxic lymphocytes of human subjects was investigated in vitro. ANF-III and ANF-IV increased blast transformation lymphocytes induced by a Con-A suboptimal dose. The increase of cells activity was comparable with the effect of interleukin-2, added at a dose of 50-100 unit/ml.

An equal high (85%) clinical efficacy of the drugs was found in 81 patients with varying stages of heart failure. A positive therapeutic effect was accompanied by lower heart rate, higher diuresis and natri-and kaliuresis, decreased systolic and diastolic pressures in the pulmonary artery.

Natural cytotoxicity was determined by assessing the cytotoxicity of individual effector cells in agarose. It has been shown that preincubation of lymphocytes with 10(-14)--10(-8) M of dalargin enhances the binding of NK-cells with targets and the formation of conjugates with killed targets and increases the percentage of active NK-cells.

View Article and Find Full Text PDFA study was made of the influence of hyperthermia (42 degrees C, 30 min) after irradiation with a dose of 6 Gy on the permeability of cell membranes and the rate of incorporation of 3H-thymidine into DNA of HeLa cells at the stationary stage of the culture growth. Hyperthermia was shown to enhance the damaging effect of radiation on cells at this growth stage. It is suggested that this effect is connected with the impaired development of a series of early (during the first minutes and hours) adaptive reactions of HeLa cells, at the stationary stage of growth, to the effect of radiation.

There were hemorrhages in the subcutaneous tissue and the muscles, kidneys, abomasum, spleen, and heart as well as liver dystrophia and hyperemia and edema of the leptomeninges and the brain cortex. Histologically, there were hyperemia, strong and diffuse pericellular and perivascular edema of the cerebrum and the cerebellum with degenerative changes in the Purkinje layer, fatty degeneration and biliary thrombi in the liver, hemorrhages and activation of the endothelial cells and glomeruli of the kidneys, hyperemia and fatty infiltration of the myocardium, and hemorrhages in the spleen.

View Article and Find Full Text PDFUsing the HeLa cell culture, the dynamics of the appearance of the mitotic block after irradiation with 600 rads has been studied, in addition to the influence of some proliferating stimulatory factors on this process. It has been shown that in the early periods after irradiation with 600 rads, on the background of the developing G2--M block, the low dose irradiation (20 rads), changes to the medium, and high temperature (41--42 degrees C) effects lead to the increase in the number of mitoses. It is supposed that this increase in the mitotic activity may be due to a heterogeneity of cell populations as to their velocity of passing through the G2-phase.
