Article Synopsis
  • The research focuses on finding effective agents against coronaviruses, specifically by synthesizing thiazolo-thiophenes based on usnic acid to inhibit SARS-CoV-2's main protease.
  • Certain modifications of the thiophene groups showed moderate antiviral activity, while others had no effect, with kinetic parameters evaluated for the most promising compound.
  • The most active compound exhibited strong binding to the protease and effective antiviral activity against various SARS-CoV-2 strains, aligning molecular dynamics results with experimental findings.

Article Synopsis
  • Tyrosyl-DNA phosphodiesterase 1 (Tdp1) is a DNA repair enzyme linked to tumor resistance against topoisomerase 1 inhibitors like topotecan.
  • Researchers synthesized new usnic acid derivatives that inhibit Tdp1 and enhance the effectiveness of topotecan in various cell lines.
  • One promising compound not only improves the response of certain mouse tumors to topotecan but also helps restore normal blood conditions in tumor-bearing mice, suggesting potential for a new cancer therapy class.

Tyrosyl-DNA phosphodiesterase 1 and 2 (Tdp1 and Tdp2) are DNA repair enzymes that repair DNA damage caused by various agents, including anticancer drugs. Thus, these enzymes resist anticancer therapy and could be the reason for resistance to such widely used drugs such as topotecan and etoposide. In the present work, we found compounds capable of inhibiting both enzymes among derivatives of (-)-usnic acid.

Article Synopsis
  • * The study assessed how these derivatives inhibit the main protease of SARS-CoV-2 and their effectiveness against a pseudoviral system showcasing the virus's glycoprotein S.
  • * Findings indicated that usnic acid and its derivatives bind to a specific site on the glycoprotein S, suggesting a potential mechanism for their antiviral action.

Tyrosyl-DNA phosphodiesterase 1 (TDP1) catalyzes the cleavage of the phosphodiester bond between the tyrosine residue of topoisomerase 1 (TOP1) and the 3' phosphate of DNA in the single-strand break generated by TOP1. TDP1 promotes the cleavage of the stable DNA-TOP1 complexes with the TOP1 inhibitor topotecan, which is a clinically used anticancer drug. This article reports the synthesis and study of usnic acid thioether and sulfoxide derivatives that efficiently suppress TDP1 activity, with IC values in the 1.

Article Synopsis
  • * Researchers synthesized ten new UA derivatives, discovering that four of them effectively inhibited TDP1 and showed low toxicity in human cell lines, while also enhancing the effectiveness of the anti-cancer drug topotecan in cancer cells.
  • * The findings suggest a potential for combining UA derivatives with topotecan in cancer treatment, but further research is needed to understand how these compounds affect non-cancerous cells differently.

The particle size distribution significantly affects the material properties of the additively manufactured parts. In this work, the influence of bimodal powder containing nano- and micro-scale particles on microstructure and materials properties is studied. Moreover, to study the effect of the protective atmosphere, the test samples were additively manufactured from 316L stainless steel powder in argon and nitrogen.

Usnic acid, a lichen secondary metabolite produced by a whole number of lichens, has attracted the interest of researchers owing to its broad range of biological activity, including antiviral, antibiotic, anticancer properties, and it possessing a certain toxicity. The synthesis of new usnic acid derivatives and the investigation of their biological activity may lead to the discovery of compounds with better pharmacological and toxicity profiles. In this context, a series of new usnic acid derivatives comprising a terpenoid moiety were synthesized, and their ability to inhibit the catalytic activity of the human DNA repair enzyme tyrosyl-DNA phosphodiesterase 1 was investigated.
