Sleep apnea (SA) is prevalent among patients with heart failure (HF) and contributes to a poor prognosis. Sodium-glucose cotransporter 2 (SGLT2) inhibitors have demonstrated efficacy in reducing the risk of serious clinical events in patients with HF. Additionally, SGLT2 inhibitors may reduce the risk of incident SA and mitigate its severity in patients with cardiovascular disease and T2DM.

The promising diagnostic performance of molecular testing for syphilis using saliva and urine samples has been reported; however, further evaluation of its possible application for diagnosis and molecular surveillance is required. In addition, the development of a rapid and easy-to-perform molecular test for syphilis is important for its use in the clinical setting. We comprehensively evaluated the diagnostic and surveillance performance of two novel loop-mediated isothermal amplification (LAMP) assays using saliva and urine samples.

Article Synopsis
  • SCLC has traditionally been viewed as a single entity, hindering advances in treatment; however, identifying genetic differences is key to improving patient outcomes.
  • A study analyzed over 1000 SCLC samples using genomic screening to identify five distinct genetic subgroups, which can respond differently to therapies, especially targeted treatments.
  • The findings indicate that while certain subgroups, like the NSCLC and MYC subgroups, have poorer survival rates with standard treatments, others may benefit from specialized therapies, highlighting the importance of personalized medicine in SCLC management.

Purpose: HER2-targeted therapies in ERBB2-amplified metastatic colorectal cancer (mCRC) are effective; however, a notable portion of patients do not respond to treatment, and secondary resistance occurs in most patients receiving these treatments. The purpose of this study was to investigate determinants of treatment efficacy and resistance in patients with ERBB2-amplified mCRC who received HER2-targeted therapy by analyzing multiomics data.

Experimental Design: We investigated genomic data from a nationwide large cancer genomic screening project, the SCRUM-Japan project.

Article Synopsis
  • The HERALD/EPOC1806 trial evaluated trastuzumab deruxtecan (T-DXd) for patients with progressive solid tumors that were identified as HER2-amplified through cell-free DNA (cfDNA) testing rather than traditional methods.
  • A total of 4,734 patients underwent cfDNA testing, with 252 showing HER2 amplification; the study ultimately included 62 patients from various cancer types, who received T-DXd treatment every three weeks.
  • The trial reported a 56.5% overall response rate (ORR), with a median progression-free survival of 7 months and noted interstitial lung diseases in 26% of patients, primarily mild to moderate cases.

Article Synopsis
  • A trial studying preoperative chemoradiotherapy (CRT) followed by nivolumab for locally advanced rectal cancer (LARC) reported promising 3-year outcomes with 39 microsatellite stable (MSS) and 5 microsatellite instability-high (MSI-H) patients.
  • For the MSS group, the 3-year relapse-free survival (RFS) rate was 79.5%, while the MSI-H group had a remarkable 100% RFS rate; overall survival (OS) rates were similarly high for both groups.
  • Key biomarkers like PD-L1 positivity and a high CD8+ T cell/effector regulatory T cell ratio appeared to contribute to better outcomes in MSS patients, highlighting potential prognostic indicators

Background And Aims: Endoscopic resection is a minimally invasive treatment for superficial esophageal squamous cell carcinoma (SESCC). Post-endoscopic resection scars complicate en-bloc resection, even with advanced techniques, such as endoscopic submucosal dissection. The cryoballoon ablation system (CBAS) effectively manages Barrett's esophagus but has limited evidence in SESCC treatment, particularly on post-endoscopic resection scars.

Interleukin-6 (IL-6)/signal transducer and activator of transcription 3 (STAT3) pathway inhibition may overcome chemoresistance of metastatic pancreatic cancer (MPC). We sought to determine the safety and recommended dose of tocilizumab (TCZ), an IL-6 receptor monoclonal antibody, and biological correlates of tumor shrinkage in patients with gemcitabine (GEM)/nanoparticle albumin-bound paclitaxel (nab-PTX)-refractory MPC. This phase 1 study enrolled 10 patients with MPC who had progressed after GEM/nab-PTX.

Serum uric acid (UA) level is associated with the high cumulative incidence or prevalence of coronary artery disease (CAD), and hyperuricemia is considered as an independent risk marker for CAD. Sleep-disordered breathing (SDB) is also associated with an increased risk of CAD. Several studies have shown that SDB is associated with hyperuricemia, but the mechanisms are unclear.

Membranes are important sites of intermolecular interactions in biological systems. However, they present significant analytical challenges as they contain multiple analytes and are dynamic in nature. In this work, we show how a Jasco J-1500 circular dichroism spectropolarimeter can be used with a microvolume Couette flow cell and appropriate cut-off filters to measure excitation fluorescence detected linear dichroism (FDLD) of fluorophores embedded in liposomal membranes.
