Cardiac macrophage contributes to the development of cardiac fibrosis, but factors that regulate cardiac macrophages transition and activation during this process remains elusive. Here we show, by single-cell transcriptomics, lineage tracing and parabiosis, that cardiac macrophages from circulating monocytes preferentially commit to macrophage-to-myofibroblast transition (MMT) under angiotensin II (Ang II)-induced hypertension, with accompanying increased expression of the RNA N6-methyladenosine demethylases, ALKBH5. Meanwhile, macrophage-specific knockout of ALKBH5 inhibits Ang II-induced MMT, and subsequently ameliorates cardiac fibrosis and dysfunction.

Although the immune checkpoint blockade (ICB) has made a great success in cancer immunotherapy, the overall response rate to the ICB, such as anti-programmed death ligand 1 (PD-L1) therapy, remains only at 20-30%. In order to improve robust operating performance and enhance bus voltage stability, a learning observer-based fault-tolerant control strategy is proposed for the distributed generation in islanded microgrid with sensor faults and uncertain disturbances. Firstly, the output feedback control theory and the linear matrix inequality method are used to design closed-loop controller for the voltage source inverter of distributed generation; secondly, a fault-tolerant model and control structure of the distributed generation in an islanded microgrid with sensor faults is analyzed. By employing the fault output signal conversion filter and proportional derivative type learning observer, the online estimation and real-time compensation of the sensor fault signal are realized.
