Collection and processing of whole blood samples in a non-clinical setting offers a unique opportunity to evaluate community-dwelling individuals both with and without preexisting conditions. Rapid processing of these samples is essential to avoid degradation of key cellular components. Included here are methods for simultaneous peripheral blood mononuclear cell (PBMC), DNA, RNA and serum isolation from a single blood draw performed in the homes of consenting participants across a metropolitan area, with processing initiated within 2 hr of collection.

DICER1 is an enzyme that generates mature microRNAs (miRNAs), which regulate gene expression post-transcriptionally in brain and other tissues and is involved in synaptic maturation and plasticity. Here, through genome-wide differential gene expression survey of post-traumatic stress disorder (PTSD) with comorbid depression (PTSD&Dep), we find that blood DICER1 expression is significantly reduced in cases versus controls, and replicate this in two independent cohorts. Our follow-up studies find that lower blood DICER1 expression is significantly associated with increased amygdala activation to fearful stimuli, a neural correlate for PTSD.

The epigenetics of early commitment to embryonal cardiomyocyte is poorly understood. In this work, we compared the effect of thyroid hormone and that of anacardic acid, a naturally occurring histone acetylase inhibitor, or both in combination, on mouse embryonic stem cells (mES) differentiating into embryonal cardiomyocyte by embryoid bodies (EBs) formation. Although the results indicated that anacardic acid (AA) and thyroid hormone were both efficient in promoting cardiomyocyte differentiation, we noticed that a transient exposure of mES to AA alone was sufficient to enlarge the beating areas of EBs compared to those of untreated controls.

Objectives: This study sought to evaluate the impact of arterial duct (AD) stenting in promoting catch-up growth of hypoplastic pulmonary artery (PA) tree in congenital heart disease with duct-dependent pulmonary circulation (CHD-DPC).

We sought to compare the abilities of the specialist Lepidoptera Pyrrhopyge thericles (Hesperiidae) and the generalist Periphoba arcaei (Saturniidae) to assimilate three highly cytotoxic compounds from their larval host plant, Vismia baccifera (Clusiaceae) and to determine whether either insect discriminated in its assimilation of the compounds that are structurally similar but of variable cytotoxicity. Objective: To examine the association between cardiometabolic risk (percent body fat [BF%], triglycerides [TG], high-density lipoprotein [HDL]-cholesterol, hemoglobin A1c [HbA1c]), socio-psychological factors (education and self-rated health [SRH]), and trajectories of grip strength (GS) in older adults.

Background: In sum, 559 Michigan schools were closed as a nonpharmaceutical intervention during the influenza A 2009 (H1N1) pandemic.

Methods: By linking the proportion of schools closed within a district to state influenza-like illness (ILI) surveillance data, we measured its effect on community levels of ILI. This analysis was centered by the peak week of ILI for each school district, and a negative binomial model compared three levels of school closure: 0%, 1%-50%, and 51%-100% of schools closed from three weeks leading up to ILI peak to four weeks following ILI peak rate.
