This review discusses the rationale for the observed regio- and stereoselectivity and the methods developed in recent years used to control and improve the stereo- and regioselectivity for the synthesis of 1,2-oxazine scaffolds.

The pathway for KDO biosynthesis has been identified as a potential target of antibacterial drug design. The reaction catalysed by KDO8P synthase is an aldol-like condensation between phosphoenolpyruvate (PEP) and d-arabinose 5-phosphate (A5P) and proceeds through a bisphosphorylated tetrahedral intermediate.
